摘 要:为筛选可用于华中樱桃遗传多样性分析和基因分型的叶绿体InDel标记,本研究以湖南省5个地理位置的华中樱桃群体为材料,鉴定了候选叶绿体非编码区序列中的InDel位点并应用于群体遗传多样性分析。结果表明,9个叶绿体非编码区片段中共有20个InDel位点,其中单碱基InDel位点9个,多碱基InDel位点11个,InDel出现频率约0.74%。非编码区trnR-UCU-atpA的InDel位点最多(5个),其次是trnHGUG-psbA (3个)和psbC-trnS-UGA (3个)。50个华中樱桃个体的核苷酸多态性(Pi)为0.001 51,可分为13个单倍型。群体ZF的核苷酸多样性最为丰富(Pi=0.001 58),其次是群体XP (Pi=0.001 40)、PJ (Pi=0.001 36)和LY (Pi=0.000 54)。从分布情况来看,单倍型H3为群体LY和PJ所共有,H5为群体ZF和XP共有,H6为群体PJ、XP和ZF共有,H8为群体PJ和XP共有;其他单倍型特异性地分布于各群体。系统进化分析结果显示,13个单倍型可分为两支:单倍型H5、H7和H11聚为一支,另外10个单倍型聚为一支,其中单倍型H5和H11以及单倍型H1、H2和H4的亲缘关系最近。本研究鉴定了20个叶绿体InDel标记,并对湖南省华中樱桃进行了基因分型和遗传多样性分析,为野生资源的保护和利用提供了理论依据。In order to screen chloroplast InDel markers that can be used for genetic diversity analysis and genotyping of Cerasus conradinae,five populations from different geographical location in Hunan Province were used as materials.The InDel loci in the candidate chloroplast non-coding region were identified and applied for population genetic diversity analysis.The results showed that there were 20 InDels in the nine chloroplast non-coding regions,including nine single-basepair InDels and 11 multi-basepair InDels.The frequency of InDels was about 0.74%.The non-coding region trnR-UCU-atpA has the most InDels(5),followed by trnH-GUG-psbA(3)and psbCtrnS-UGA(3).The nucleotide diversity(Pi)of 50 individuals of C.conradinae is 0.00151,which can be divided into 13 haplotypes.Population ZF has the most abundant nucleotide diversity(Pi=0.00158),followed by population XP(Pi=0.00140),PJ(Pi=0.00136)and LY(Pi=0.00054).From the distribution point of view,haplotype H3 is shared by populations LY and PJ;H5 is shared by populations ZF and XP;H6 is shared by populations PJ,XP and ZF;H8 is shared by populations PJ and XP.Other haplotypes are geographically distributed in a specific population.The results of systematic evolution analysis showed that the 13 haplotypes could be divided into two clades:haplotypes H5,H7 and H11 were clustered in clade one,and the other 10 haplotypes were clustered in clade two.The closest relationship was observed between haplotype H5 and H11 and also among haplotypes H1,H2 and H4.In this study,20 chloroplast InDel markers were identified,and the genotyping and genetic diversity analysis of C.conradinae in Hunan Province were carried out,which provided a theoretical basis for the protection and utilization of wild resources.
