摘 要:为建立羊耳菊总酚的含量测定方法,考察云南不同产地羊耳菊总酚含量差异,通过单因素试验和正交试验优化提取工艺,以麝香草酚为对照品,采用可见分光光度法测定羊耳菊的总酚含量。研究结果表明:对照品麝香草酚溶液在波长278 nm处有最大吸收,在9.8~68.6 mg/L范围内浓度与吸光度呈良好的线性关系(r^(2)=0.9992),方法精密度RSD为1.45%,加标平均回收率为98.62%,分析方法稳定可靠。9批不同产地羊耳菊总酚的质量分数有一定差异,总体在7.61~2.89 mg/g之间。实验结果可为提高羊耳菊药材的质量标准,进一步开发利用羊耳菊产品提供一定的参考。A method for determination of the total phenolic content of Inula cappa was established,and the amount of total phenol in Inula cappa from different production areas in Yunnan was compared.The extraction technology was optimized through single factor test and orthogonal testand the content of total phenolin Inula cappa was determined by visible spectrophotometry with thymol as control substance.The results showed that the maximum absorbance of thymol solution was obtained at the wavelength of 278 nm,and there was a good linear relationship between the concentration and absorbance in the range of 9.8~68.6 mg/L(r^(2)=0.9992).The precision RSD of the method was 1.45%,and the average recovery rate was 98.62%.The analysis method was stable and reliable.The content of total phenol in 9 batches from different producing areas was different,ranging from 7.61~2.89 mg/g.The experimental results can provide some reference for improving the quality standard and further developing and utilizing the products of Inula cappa.
