摘 要:文章建立一种邻苯二甲醛(OPA)柱前衍生-高效液相色谱检测构树饲料中的氨基酸方法。采用酸水解样品,研究氮气通入、水解温度、水解时间、水解液浓度对水解的影响,找到最佳的水解条件;样品水解后,用OPA柱前衍生进行HPLC分析。结果表明,水解的最优条件为:通入氮气作保护气,用6mol/L的HCl溶液在110℃下水解24 h。杂交构树饲料的总氨基酸为8.53 g/100 g;野生构树饲料的总氨基酸为7.11 g/100 g,与常用的玉米饲料氨基酸总量相当。该方法操作简单、分析快速、适用范围广、灵敏度高,为构树饲料中的成分检测及开发利用提供了理论依据。A method was developed for the determination of amino acids in the feeds of broussonetia papyrifera by HPLC using o-phthalaldehyde(OPA)as derivatizer.The optimal hydrolysis conditions were found by investigating the influences of nitrogen introduction,hydrolysis temperature,hydrolysis time and extraction acid concentration on the hydrolysis.The optimum conditions for acid hydrolysis were as follows:nitrogen gas as protective gas,6 mol/L hydrochloric acid solution as hydrolysis solution,hydrolysis time of 24 h and hydrolysis temperature of 110℃.The total amino acid content of hybrid broussonetia papyrifera feed was 8.53 g/100 g.The total amino acid content of wild broussonetia papyrifera feed was 7.11 g/100 g,which was not much different from that of the commonly used corn feed.The method is simple,rapid,wide applicability and high sensitivity,which provides a theoretical basis for the detection and development of the components in broussonetia papyrifera feed.
