摘  要:研究了17、25和33℃3个干露温度对拟穴青蟹幼蟹存活、血蓝蛋白和细胞凋亡相关基因表达的影响.结果发现,17℃组拟穴青蟹幼蟹存活率最高为58.93%,且存活时间最长为144 h;33℃组存活率最低为17.86%,且存活时间最短为120 h.17、25和33℃组的血蓝蛋白分别在48、96、48 h达到峰值,25℃和33℃组的皮质醇含量在96 h时显著高于其他时间点(P<0.01),而活性氧ROS含量随干露时间增加而上升.在基因表达方面,干露时间增加导致bax基因和bcl-2基因上调,其中17℃和33℃组在干露24 h时的bax/bcl-2比值大于1,且显著高于其他时间点(P<0.05),caspase-3基因表达量在17℃和33℃组干露24 h时也显著高于其他时间点(P<0.01).双因素方差分析表明,温度、干露及其两者的交互作用均显著影响了拟穴青蟹幼蟹的存活率(P<0.01).以上研究结果表明,温度和干露胁迫对拟穴青蟹幼蟹存活的影响可能是通过应激反应和凋亡途径介导的,17℃是拟穴青蟹幼蟹最适宜的运输温度.The effects of air-exposure temperatures(17,25 and 33℃)on the survival and apoptosis-related gene expression of juvenile mud crab Scylla paramamosain were investigated.The results showed that the 17℃group had the highest survival rate,while the 33℃group had the lowest survival rate.Hemocyanin in 17,25 and 33℃groups peaked at 48,96,and 48 h,respectively.Cortisol content in 25℃and 33℃groups was the highest at 96 h(P<0.01);whereas reactive oxygen species(ROS)content increased with extending air-exposure duration.In terms of gene expression,bax and bcl-2 genes were significantly up-regulated with increasing air-exposure duration.Two-way analysis of variance showed that temperature,air-exposure and their interaction significantly affected the survival rate of juvenile mud crab(P<0.01).Based on the above research results,the effects of temperature and air-exposure on the survival of juvenile mud crab may be mediated by stress response and apoptosis pathway.In addition,the optimum temperature for the transportation of juvenile mud crab was 17℃.
