血清lncRNA GAS5表达与结直肠癌血管生成的关系及其临床意义

摘　　要：[目的]检测结直肠癌(colorectal cancer,CRC)患者血清长链非编码RNA生长停滞特异基因5(lncRNA GAS5)表达量,探讨其与肿瘤血管生成的关系及其临床意义。[方法]以实时荧光定量PCR检测90例CRC患者(CRC组)术前、30位健康者(正常对照组)血清lncRNA GAS5表达量,以ELSIA法检测血清血管内皮生长因子(VEGFA)含量;以免疫组化法检测CRC组及正常对照组组织中CD34表达,计算肿瘤微血管密度(microvessel density,MVD);随访CRC患者术后预后3年;分析术前血清lncRNA GAS5表达量与CRC血管生成、病理特征及预后的关系。[结果]CRC组血清lncRNA GAS5表达量显著低于正常对照组(P<0.01),血清VEGFA含量及癌组织MVD显著高于正常对照组(P<0.01);lncRNA GAS5低表达者血清VEGFA含量及MVD均显著高于lncRNA GAS5高表达者(P<0.01);CRC组血清lncRNA GAS5表达量与血清VEGFA含量及MVD均呈负相关;术前血清lncRNA GAS5表达量与CRC分化程度、TNM分期、淋巴结转移及术后复发转移呈负相关(P<0.01),与术后生存率呈正相关(P<0.01)。[结论]血清lncRNA GAS5表达量降低可促进CRC新生血管生成及侵袭转移,与CRC不良预后相关;术前血清lncRNA GAS5表达量可成为CRC预后评价指标。[Objective]To investigate the correlation between the expression levels of long non-coding RNA growth arrest specific gene 5(lncRNA GAS5)in serum and angiogenesis in CRC and its clinical significance.[Methods]The expression levels of lncRNA GAS5 in serum weredetected by real-time PCR,the VEGFA in serum were detected by ELISA.The expression of CD34 were detected by immunohistochemistry in CRC(n=90)and normal control(n=30).The MVD were calculated.The 3 years postoperative prognosis was followed-up.The relationship between the preoperative serosal lncRNA GAS5 expression levels in the CRC and angiogenesis,the pathologic features and prognosis were analyzed.[Results]The serum lncRNA GAS5 expression levels in CRC group were significantly lower than those in the normal control group(P<0.01),the VEGFA serum levels and MVD in CRC group were significantly higher than those in the normal control group(P<0.01).The VEGFA serum expression levels and MVD in low lncRNA GAS5 expression group were significantly higher than those in high lncRNA GAS5 expression group.The serum lncRNA GAS5 expression levels were negatively related with the serum VEGFA expression levels and MVD in CRC group.The preoperative serum lncRNA GAS5 expression levels in CRC group were negatively related with the differentiated degree of tumor,TNM stage,lymph node metastasis and postoperative recurrence and metastasis(P<0.01),and were positively related with the postoperative survival(P<0.01).[Conclusion]The low serum lncRNA GAS5 expression levels in CRC promote tumor angiogenesis and invasion and metastasis,were related with poor prognosis,and can be a prognostic indicator of CRC.
