摘 要:目的:利用酶解法制备夏桑菊低聚糖,考察其对益生菌的增殖作用,并优化酶解工艺。方法:分别考察8种不同生物酶制备的夏桑菊低聚糖对5种不同益生菌生长活性,选出增殖率最高的酶与益生菌组合,用于酶解工艺优化;采用单因素设计方法,考察酶添加量、酶解温度、酶解时间工艺参数对益生菌增殖率的影响。结果:利用葡聚糖酶制备的夏桑菊低聚糖对鼠李糖乳杆菌的促增殖率最高,增殖率为(265±14.5)%。酶添加量为1500 U/mL时,鼠李糖乳杆菌的增殖率最高,达到了(320±17.2)%;酶解温度50℃时,鼠李糖乳杆菌增殖效果最高,增殖率为(329±15.6)%;酶解时间为4 h时,鼠李糖乳杆菌的增殖率最高,增殖率为(364±16.4)%。结论:8种不同生物酶酶解制备的夏桑菊低聚糖对5种益生菌都具有较好的促增殖作用,其中利用葡聚糖酶制备的夏桑菊低聚糖对鼠李糖乳杆菌的增殖效果最好,从益生活性考虑,选取葡聚糖酶添加量1500 U/mL、酶解温度50℃和酶解时间4 h为较优酶解方案。Objective To prepare Xiasangju oligosaccharides by enzymolysis method,investigate its effect on the proliferation of probiotics,and optimize the enzymolysis process.Methods The growth activities of the oligosaccharides prepared by 8 kinds of different enzymes on 5 kinds of different probiotics were investigated,and the combination of enzyme and probiotics with the highest additive rate was selected to optimize the enzymatic hydrolysis process.The single factor design method was used to investigate the effects of enzyme addition amount,enzymatic hydrolysis temperature and enzymatic hydrolysis time on the additive rate of probiotics.Results The oligosaccharides prepared by glucanase had the highest proliferation rate against Lactobacillus rhamnosus,and the proliferation rate was(265±14.5)%.When the enzyme dosage was 1500 U/mL,the proliferation rate of Lactobacillus rhamnosus was the highest,reaching(320±17.2)%.When the enzymatic hydrolysis temperature was 50℃,the proliferation effect of Lactobacillus rhamnosus was the highest,and the proliferation rate was(329±15.6)%.When the enzymolysis time was 4 h,the proliferation rate of Lactobacillus rhamnosus was the highest(364±16.4)%.Conclusion The oligosaccharides prepared by enzymolysis of 8 different bioenzymes had a good effect on the proliferation of 5 kinds of probiotics,among which the oligosaccharides prepared by glucanase had the best effect on the proliferation of Lactobacillus rhamnosus.Considering the probiotic activity,the optimal solution was selected as the dosage of glucanase 1500 U/mL,enzymolysis temperature 50℃and enzymolysis time 4 h.
