摘  要:目的通过体外实验水飞蓟素处理弥漫大B细胞淋巴瘤细胞来研究其对细胞增殖和凋亡的影响。方法使用CCK-8方法分析水飞蓟素抑制弥漫大B细胞淋巴瘤细胞株OCI-LY-19的增殖活性;通过Annexin V-FITC/PI实验分析水飞蓟素诱导细胞凋亡率。通过实时定量PCR和Western blotting实验检测水飞蓟素作用后AKT和Caspase-3基因和蛋白质水平的变化。结果与0 ng/ml水飞蓟素相比,10、20、50、100 ng/ml水飞蓟素的细胞增殖活性抑制率较高,差异有统计学意义(P<0.05)。水飞蓟素作用OCI-LY-19细胞48 h的半数抑制浓度(IC50)为73.27 ng/ml。经50、100、150 ng/ml浓度的水飞蓟素作用48 h后,早期和晚期细胞凋亡比例总和相对于0 ng/ml组逐渐升高,差异有统计学意义(P<0.05)。随着0~150 ng/ml药物浓度的递增,OCI-LY-19细胞中AKT mRNA表达水平逐渐下降,Caspase-3 mRNA表达水平逐渐升高,各组差异均有统计学意义(P<0.05)。与对照组0 ng/ml比较,随着水飞蓟素浓度升高,细胞中AKT蛋白水平逐渐下降,Caspase-3蛋白水平逐渐升高,各组差异均有统计学意义(P<0.05)。结论水飞蓟素可能通过调节PI3K/AKT/Caspase-3途径来发挥对人弥漫大B细胞淋巴瘤OCI-LY-19细胞的抗肿瘤作用。Objective To study the effect of silymarin on the proliferation and apoptosis of OCI-LY-19 cells in diffuse large B lymphoma by experiment in vitro.Methods The OCI-LY-19 cells were treated with silymarin and the inhibitory effect on proliferation was detected by CCK-8 method.The apoptosis rate of cells after silymarin was analyzed by Annexin V-FITC/PI assay.Quantitative real time PCR and Western blotting methods were used to detect mRNA and protein levels of AKT and Caspase-3 in cells treated with silymarin.Results Compared with 0 ng/ml,the OCI-LY-19 cells in 10,20,50 and 100 ng/ml silymarin groups showed higher inhibitory rate of proliferation activity,with statistical significances(P<0.05).The median inhibitory concentration(IC50)of silymarin treated OCI-LY 19 cells for 48 h was 73.27 ng/ml.After treatment with 50,100,150 ng/ml of silymarin for 48 h,the sum proportion of early and late apoptotic cells increased gradually compared with 0 ng/ml group,the differences were statistically significant(P<0.05).With the increase of drug concentration from 0 to 150 ng/ml,the mRNA expression level of AKT in OCI-LY-19 cells was gradually decreased,and the level of Caspase-3 was gradually increased,with statistical significances among all groups(P<0.05).Compared with the control group at 0 ng/ml,with the increase of silymarin concentration,the level of AKT protein in cells gradually decreased and the level of Caspase-3 gradually increased,with statistical significances among all groups(P<0.05).Conclusion Silymarin may exert its antitumor effect on human diffuse large B-cell lymphoma OCI-LY-19 cells by regulating the PI3K/AKT/Caspase-3 pathway.
