摘  要:WRKY转录因子是目前研究的与逆境有关的转录因子之一。本试验以马铃薯栽培品种‘冀张薯12号’为材料,获得StWRKY6基因,并对其进行了生物信息学分析和功能的初步研究。StWRKY6基因长度为1491 bp,由496个氨基酸编码而成,其相对分子量为55212.93,理论等电点为7.02,带负电残基总数为48,带正电残基总数为47,不稳定系数为51.70。该蛋白质二级结构以无规则卷曲为主,占54.9%,α-螺旋为32.3%,延伸链为9.7%,β-折叠为3.2%,是不稳定蛋白,属于亲水蛋白。通过进化树分析,与番茄同源性最高,可达90%以上;利用RT-qPCR构建表达谱,发现StWRKY6在叶片中的表达量较高;经过低温胁迫和盐胁迫处理一定时间后其表达量显著增加,说明其可能与马铃薯的耐盐性和耐低温性有关。本研究为深入研究马铃薯StWRKY转录因子参与非生物胁迫调控提供科学依据。WRKY transcription factor is one of the transcription factors related to stress.In this experiment,the StWRKY6 gene was obtained from'Jizhangshu 12',a potato cultivar,and its bioinformatics analysis and function were studied.The results showed that the length of StWRKY6 gene was 1491 bp,which was encoded 496 amino acids.Its molecular weight was 55212.93,theoretical isoelectric point was 7.02,total number of negatively charged residues were 48,total number of positively charged random coil 47,and instability coefficient was 51.70.The secondary structure of the protein was mainly random curve,accounting for 54.9%,α-helix 32.3%,extended strand 9.7%,β-sheet 3.2%.WRKY6 protein was unstable and belongs to hydrophilic protein.Phylogenetic tree analysis showed that StWRKY6 had the highest homology with tomato(more than 90%);RT-qPCR analysis showed that the expression level of StWRKY6 gene was the highest in leaves;after being treated with low temperature stress and salt stress for a certain period of time,its expression level increased significantly,which indicating that StWRKY6 gene may be related to salt tolerance and low temperature tolerance of potato.This study provides a scientific basis for in-depth study of potato StWRKY transcription factor involved in abiotic stress regulation.
