摘 要:丁香酸是一种植物酚酸化合物,因其抗氧化、降脂和保肝作用而受到关注。本研究旨在探索丁香酸联合游泳对NAFLD大鼠肝功能的效果。本研究将SD大鼠被分为7组,每组12只:CON组、NAFLD组、SWI组、L-SA组、M-SA组、H-SA组及SWI+H-SA组。CON组大鼠被给予正常饲料,其他组则被喂食70%的高脂饲料。经过12周后,SWI组开始游泳训练。L-SA、M-SA和H-SA组的大鼠分别灌胃2 m L 25、50和100 mg·kg-1·d-1的丁香酸溶液,而SWI+H-SA组同时进行游泳与灌胃治疗。控制组则灌胃2 mL的生理盐水,灌胃治疗持续8周。通过各种生化检测手段,观察了大鼠的体重变化、血清生化指标、肝脏氧化应激水平,并利用RT-qPCR技术检测了肝脏中的脂质代谢关键基因的转录活性,同时使用Western blot技术研究了Nrf2和AMPK在肝脏中的激活情况。结果显示,与SWI组及H-SA组相比,SWI+H-SA组大鼠体重明显降低,相关血清生化指标如ALT、AST、TC、TG、LDL-c及FFA均有所下降,而HDL-c则上升。此外,肝脏形态、抗氧化酶活性和关键脂代谢基因的表达也出现了有利变化,尤其是Nrf2和AMPKα的活化显著增强。本研究证明,丁香酸结合游泳能够显著改善NAFLD大鼠的肝脏功能,这种保护作用可能与其对氧化应激的抑制、脂代谢的调节以及Nrf2和AMPK信号通路的激活有关。因此,丁香酸与游泳的联合应用可能为NAFLD的综合治疗提供新的策略。Syringic acid is a plant phenolic compound that has garnered attention for its antioxidative,lipid-lowering,and hepato-protective properties.This study aimed to explore the effects of the combined treatment of syringic acid and swimming on the liver function of NAFLD rats.In this study,SD rats were divided into seven groups with12 rats in each group:CON group,NAFLD group,SWI group,L-SA group,M-SA group,H-SA group,and SWI+H-SA group.Rats in the CON group were fed with normal feed,while the other groups received a diet containing 70%high fat.After 12 weeks,the SWI group started swimming training.The L-SA,M-SA,and H-SA groups were respectively gavaged with 2 mL of syringic acid solution at concentrations of 25,50,and 100 mg·kg~(-1)·d~(-1).The SWI+H-SA group underwent both swimming and gavage treatments simultaneously.The control group was gavaged with 2 mL of saline,continuing this for 8 weeks.Various biochemical assays were conducted to monitor changes in rat weight,serum biochemical indicators,and liver oxidative stress levels.The RT-qPCR technique was utilized to detect the transcriptional activity of key lipid metabolism genes in the liver.In addition,the Western blot technique was used to study the activation of Nrf2 and AMPK in the liver.Results indicated that,compared to the SWI and H-SA groups,the SWI+H-SA group exhibited a significant reduction in rat weight and a decrease in serum biochemical markers such as ALT,AST,TC,TG,LDL-c,and FFA,whereas HDL-c increased.Moreover,liver morphology,antioxidative enzyme activities,and the expression of key lipid metabolism genes exhibited beneficial changes,especially the marked enhancement of Nrf2 and AMPKαactivation.The findings suggest that the combination of syringic acid and swimming notably improves liver function in NAFLD rats.This protective effect might be related to its inhibition of oxidative stress,regulation of lipid metabolism,and activation of the Nrf2 and AMPK signaling pathways.Therefore,the combined application of syringic acid and swimming may provi
