摘　　要：目的探讨miR-183在HPV相关型宫颈腺癌中的表达及与富含亮氨酸重复序列免疫球蛋白样结构域基因1(LRIG1)的相互作用关系。方法实时定量PCR法检测HPV相关型宫颈腺癌组织中miR-183及LRIG1 mRNA的表达水平,Western blot法检测腺癌组织中LRIG1蛋白的表达。将miR-183 inhibitors(抑制物)、miR-183 mimics(模拟物)应用阳离子脂质体Lipofectamine TM2000转染至宫颈腺癌Hela细胞,并设立空白对照组、各自的阴性对照组,采用实时定量PCR法检测各组细胞中miR-183及LRIG1 mRNA的表达水平,Western blot法检测宫颈各组细胞中LRIG1蛋白的表达水平。结果miR-183 mRNA在HPV相关型宫颈腺癌组织中的表达(0.91±0.10)明显高于正常宫颈组织(0.52±0.06),差异有统计学意义(P<0.05),而LRIG1的mRNA(0.56±0.03)及蛋白(0.09±0.02)表达水平均明显低于正常宫颈组织(mRNA:1.12±0.08,蛋白:0.25±0.05),差异有统计学意义(P<0.05)。与对照组(1.06±0.06)相比,转染miR-183抑制物的腺癌Hela细胞中miR-183 mRNA的表达水平(0.69±0.07)显著降低,差异有统计学意义(P<0.05),转染miR-183抑制物的腺癌Hela细胞和对照组中,LRIG1 mRNA(分别为1.75±0.10、0.71±0.09)及其蛋白(分别为1.16±0.09、0.76±0.04)的表达水平反而均明显增高,差异均有统计学意义(P<0.05);与对照组(0.99±0.05)相比,转染miR-183模拟物的宫颈腺癌Hela细胞中miR-183 mRNA的表达水平(2.55±0.25)显著增高(P<0.05),转染miR-183模拟物的腺癌Hela细胞和对照组中LRIG1 mRNA(分别为0.40±0.05、0.75±0.08)及其蛋白(分别为0.32±0.01、0.76±0.06)表达水平反而均明显降低(P<0.05)。结论miR-183在HPV相关型宫颈腺癌中呈高表达,可能通过抑制LRIG1的表达参与宫颈腺癌的发生和发展。Objective To investigate the expression of miR-183 and the interaction of miR-183 with LRIG1 in HPV-associated adenocarcinoma of cercix.Methods The expression of miR-183 and LRIG1 mRNA was detected by real-time quantitative PCR in HPV-associated adenocarcinoma of cercix tissues,and the expression of LRIG1 was detected by western blot in adenocarcinoma tissues;The miR-183 inhibitors and miR-183 mimics were transfected into Hela cells of cervical adenocarcinoma using cationic liposome Lipofectaminet M2000,and blank control group and negative control group were set up.The expression of miR-183 and LRIG1 mRNA in each group was detected by real-time quantitative PCR,the protein expression of LRIG1 was detected by western blot.Results The expression level of miR-183 was significantly higher in HPV-associated adenocarcinoma of cercix tissues(0.91±0.10)than that in normal tissues(0.52±0.06)(P<0.05).And the expression levels of LRIG1 mRNA(0.56±0.03)and protein(0.09±0.02)were significantly decreased in HPV-associated adenocarcinoma of cercix than that in normal tissues(mRNA:1.12±0.08,protein:0.25±0.05)(P<0.05).The expression level of miR-183 mRNA was significantly decreased in adenocarcinoma Hela cells transfected with miR-183 inhibitors(0.69±0.07)compared with the control group(1.06±0.06)(P<0.05),On the contrary,in Hela cells transfected with miR-183 inhibitors and control group,the expression levels of LRIG1 mRNA(1.75±0.09,0.71±0.09)and its protein(1.16±0.09,0.76±0.04)were significantly increased(P<0.05);The expression level of miR-183 mRNA in cervical adenocarcinoma Hela cellstransfected with miR-183 mimics was significantly(2.55±0.25)increased than the control group(0.99±0.05)(P<0.05),while in Hela cells transfected with miR-183 mimics and control group,the expression level of LRIG1 mRNA(0.39±0.05,0.74±0.08)and protein(0.32±0.01,0.76±0.05)was significantly decreased(P<0.05).Conclusion MiR-183 is expressed highly in HPV-associated adenocarcinoma of cercix,which may be involved in the occurrence and dev
