摘 要:开发一种利用银枝晶作为表面增强拉曼光谱基底,结合偶氮偶联反应对组氨酸进行快速灵敏检测的方法。通过分别考察3-氨基苄胺、3-苯胺磺酸和2-氟-4-巯基苯胺(2-fluoro-4-mercaptoaniline,2-F-PATP)与组氨酸偶氮偶联反应得到衍生化产物的拉曼光谱发现:2-F-PATP对组氨酸的拉曼指纹图谱效果最好,对组氨酸检出限为10^(-21)mol/L。选择组氨酸偶氮偶联反应后的偶氮产物特征峰(1387 cm^(-1)和1432 cm^(-1))进行定量分析,特征峰拉曼光谱强度与组氨酸浓度在一定范围内呈线性关系,相关系数R^(2)分别为0.97561和0.96884。本方法可用于牛血清白蛋白、人血清白蛋白、α-乳白蛋白和苹果汁等的检测。本检测方法将表面增强拉曼光谱和偶氮偶联反应结合,具有灵敏度高、操作简便的特点,有良好的应用前景。A method was developed for the rapid and sensitive detection of histidine using silver dendrimers as a substrate for surface-enhanced Raman spectroscopy in combination with azo-coupling reaction.The azo-coupling reaction of 3-aminobenzylamine,3-anilinosulfonic acid or 2-fluoro-4-mercaptoaniline(2-F-PATP)with histidine was investigated.The Raman spectra of the resulting products revealed that 2-F-PATP gave the best Raman fingerprint for histidine,with a limit of detection(LOD)of 10^(-21)mol/L.The characteristic peaks(at 1387 and 1432 cm^(-1))of the azo-coupling product of histidine were selected for quantitative analysis,and the intensity of the characteristic peaks in the Raman spectra showed a linear relationship with histidine concentration in a certain range,with correlation coefficients(R^(2))of 0.97561 and 0.96884,respectively.This method can be used for the determination of bovine serum albumin,human serum albumin,α-lactalbumin and apple juice.The proposed method is sensitive and simple to use,and has good application potential.
