摘  要:透明质酸(HA)是一种由N-乙酷葡萄糖胺(GIcNAc)和D-葡萄糖醛酸(GlcA)以β-1,3和β-1,4糖苷键交替连接组成的高聚合大分子黏性多糖,具有极强的保湿作用,被称为理想的天然保湿因子,是目前自然界中发现的化妆品用保湿性能最好的物质。马疫链球菌是目前透明质酸生产的主要菌种。菌株诱变是提升菌株性能或提高代谢产物的常用方法。采用ARTP对菌株进行诱变,通过改良CTAB比浊法进行菌株高通量初筛和摇瓶复筛,显著提高了透明质酸生产能力。经过3轮ARTP迭代诱变和筛选,最终使菌株HA产量提高了28.7%。Hyaluronan or hyaluronic acid(HA),is a glycosaminoglycan composed of repeating polymeric disaccharides of uronic sugar(D-glucuronic acid)and amino sugar(N-acetyl-D-glucosamine),both linked by glucoronidicβ-1,3 glycosidic bonds.Streptococcus equi subsp.zooepididicus is the main strain in HA production.Strain mutagenesis is a commonly used method to improve strain performance or enhance metabolic products.In this paper,ARTP was used to mutate the strain,and improved CTAB turbidimetric method was used for high flux initial screening and shake flask re-screening of the strain,significantly improving the production capacity of hyaluronic acid.After three rounds of ARTP iteration mutagenesis and screening,the HA production of the strain was ultimately increased by 28.7%.
