摘 要:目的:观察外湿对健康大鼠及胶原诱导的关节炎(CIA)模型大鼠的干预作用,探寻“湿流关节”的内涵。方法:24只SD大鼠随机分为对照组、湿邪干预对照组、CIA组、湿邪干预CIA组,每组6只。CIA组和湿邪干预CIA组造CIA模型,湿邪干预对照组与湿邪干预CIA组自分组后置于人工模拟气候箱内接受高湿度干预60d。采用0~4级关节炎评分法统计大鼠关节炎评分,番红O染色法观察大鼠关节软骨病理组织学情况,ELISA法测大鼠血清TNF-α、IL-17、COX-2、C5a含量,免疫组织化学法测大鼠关节软骨C5aR含量。结果:与对照组比较,湿邪干预对照组关节炎评分显著上升(P<0.01),关节软骨组织部分软骨细胞着色力减弱,血清中TNF-α及C5a含量显著增加(P<0.05),关节软骨中C5aR表达量显著增加(P<0.01);CIA组关节炎评分显著上升(P<0.01),关节软骨组织部分软骨细胞着色力减弱,血清中TNF-α、IL-17、COX-2、C5a含量显著增加(P<0.05),关节软骨中C5aR表达量显著增加(P<0.05)。与CIA组比较,湿邪干预CIA组关节炎评分显著上升(P<0.05),关节软骨组织软骨细胞着色力减弱,血清中TNF-α、IL-17、COX-2、C5a含量显著增加(P<0.05),关节软骨中C5aR表达量显著增加(P<0.01)。结论:外湿可直接诱发或加重关节损伤,引发“湿流关节”,其内在机制之一可能与C5a-C5aR轴驱使炎性细胞定向关节趋化相关。Objective:To observe the intervention effect of external dampness on healthy SD rats and collagen-induced arthritis(CIA)rats,and to explore the connotation of‘dampness flowing into joints'.Methods:A total of 24 SD rats were randomly divided into a control group,damp intervention control group,CIA group,and damp intervention CIA group,with 6 rats in each group.CIA group and damp intervention CIA group were treated with a collagen-induced arthritis model.The damp intervention control group and damp intervention CIA group were placed in an artificially simulated climate box to receive high humidity intervention for 60 days after grouping.The O-4 grade arthritis scoring method was used to evaluate the arthritis score of rats;the safranin O staining method was used to observe the pathological histology of rat joint cartilage;ELISA method was used to measure serum TNF-α,IL-17,COX-2,and C5a levels in rats;and immunohistochemistry was used to measure the C5aR levels in rat joint cartilage.Results:Compared with the control group,the arthritis score was significantly increased(P<0.01)the staining ability of a part of the chondrocyte of cartilage was reduced,the contents of TNF-α and C5a in serum were significantly increased(P<0.05),and the expression of C5aR in articular cartilage was significantly increased(P<0.01)in the damp intervention control group;the arthritis score of the CIA group was significantly increased(P<0.01),the staining ability of chondrocyte of articular cartilage was reduced,the contents of TNF-α,IL-17,COX-2,and C5a in serum were significantly increased(P<0.05),and the expression of C5aR in articular cartilage was significantly increased(P<0.05).Compared with the CIA group,the arthritis score of the damp intervention CIA group was significantly increased(P<0.05),the staining ability of chondrocyte of articular cartilage was reduced and the contents of TNF-α,IL-17,COX-2,and C5a in serum were significantly increased(P<0.05),the expression of C5aR in articular cartilage was significantly increased(P<
