摘 要:目的:探究KDM6A在口腔鳞状细胞癌发生发展中的作用机制。方法:用生物信息学方法分析KDM6家族成员mRNA表达量与口腔鳞状细胞癌患者恶性程度及预后的相关性,并通过组织芯片免疫组化进行验证。构建KDM6A过表达的CAL-27细胞株,分别利用CCK8、EDU、划痕修复、Transwell实验检测细胞的增殖和迁移情况,并通过RNA-seq探寻KDM6A调控口腔鳞状细胞癌的可能机制。结果:KDM6A在口腔鳞状细胞癌中低表达,与肿瘤的恶性程度及患者的预后相关;过表达KDM6A后CAL-27的增殖和迁移能力受到抑制;RNA-seq结果表明Hippo signaling pathway可能是KDM6A的下游信号通路。结论:KDM6A可能通过Hippo信号通路调控GSK3B抑制口腔鳞状细胞癌的增殖和迁移。Objective:To explore the mechanism of KDM6A in the development of oral squamous cell carcinoma(OSCC).Methods:The correlation between the expression of KDM6 family mRNA and the malignant degree and prognosis of OSCC was analyzed by bioinformatics method,and confirmed by tissue microarray immunohistochemistry.The KDM6A overexpression OSCC cell line CAL-27 was constructed,and the proliferation and migration of the cells were detected by CCK8,EDU,scratch repair and Transwell assays.The possible mechanism of KDM6A regulation in OSCC was explored by RNA-seq.Results:The low expression of KDM6A in OSCC was correlated with the malignancy and prognosis of the patients.Overexpression of KDM6A inhibited the proliferation and migration of CAL-27 cells.RNA-Seq results suggested that Hippo signaling pathway may be a downstream pathway of KDM6A.Conclusion:KDM6A may inhibit the proliferation and migration of OSCC by regulating GSK3B through Hippo signaling pathway.
