摘  要:目的 观察肺腺癌组织中赖氨酸乙酰转移酶5[K(lysine) acetyltransferase,KAT5]和鼠双微体2(Murine double minute2,MDM2)的表达变化,探讨其临床意义。方法 60例通过手术切除且经过病理学确诊肺腺癌患者,留取癌组织和癌旁肺组织。采用荧光免疫组织化学法对肺腺癌组织及癌旁组织KAT5、MDM2进行定位检测,采用免疫组织化学法对肺腺癌组织及癌旁组织KAT5、MDM2进行半定量检测,分析KAT5、MDM2阳性表达与肺腺癌患者临床病理参数的关系,采用Spearman相关检验法分析肺腺癌组织KAT5、MDM2表达的相关性。结果 相比于癌旁组织,肺腺癌组织中KAT5、MDM2高表达。肺腺癌组织及癌旁组织KAT5阳性表达率分别为65.00%(39/60)、45.00%(27/60),二者比较,P<0.05;MDM2阳性表达率分别为78.33%(47/60)、40.00%(24/60),二者比较,P<0.05。KAT5、MDM2阳性表达与肺腺癌患者的TNM分期、肿瘤分化程度和淋巴结转移密切相关(P均<0.05)。肺腺癌组织中KAT5、MDM2的表达呈正相关(r=0.632,P<0.05)。结论 肺腺癌组织中KAT5、MDM2呈高表达。KAT5、MDM2阳性表达与肺腺癌患者的TNM分期和淋巴结转移相关。KAT5、MDM2可能协同促进肺腺癌的发生发展。Objective To observe the expression changes of K(lysine)acetyltransferase(KAT5)and murine double minute 2(MDM2)in lung adenocarcinoma tissues,and to explore their clinical significance.Methods Sixty cases of patients who were pathologically confirmed as lung adenocarcinoma were selected,and their lung adenocarcinoma carcinoma and normal lung tissue were collected to detect the localization and semi-quantitation of KAT 5 and MDM 2 by fluorescence immunohistochemistry(IF)and immunohistochemistry,respectively.We analyzed the relationships between positive expression of KAT5 and MDM2 and clinicopathologic parameters of lung adenocarcinoma patients,and the correlation of KAT5 and MDM2 expression in lung adenocarcinoma tissues was analyzed by Spearman correlation test.Results The expression of KAT5 and MDM2 in lung adenocarcinoma tissues was higher than that in the adjacent tissues.The positive expression rates of KAT5 in the lung adenocarcinoma tissues and adjacent tissues were 65.00%(39/60)and 45.00%(27/60),respectively,with statistically significant difference(P<0.05).The positive expression rates of MDM2 were 78.33%(47/60)and 40.00%(24/60),respectively,with statistically significant difference(P<0.05).The positive expression levels of KAT5 and MDM2 were closely correlated with TNM stage,tumor differentiation degree and lymph node metastasis of lung adenocarcinoma patients(all P<0.05).The expression of KAT5 and MDM2 in lung adenocarcinoma tissue was positively correlated(r=0.632,P<0.05).Conclusions KAT5 and MDM2 are highly expressed in lung adenocarcinoma tissues.The positive expression levels of KAT5 and MDM2 are correlated with TNM stage and lymph node metastasis in patients with lung adenocarcinoma.KAT5 and MDM2 may synergistically promote the development and progression of lung adenocarcinoma.
