摘 要:目的:观察温肾阳颗粒对去卵巢骨质疏松模型小鼠骨量及骨强度的影响。方法:选取3个月龄SPF级C57BL/6品系雌性小鼠60只,采用随机数字表法分为正常对照组、模型组、温肾阳颗粒低剂量组、温肾阳颗粒中剂量组、温肾阳颗粒高剂量组,每组12只。正常对照组仅切除卵巢周围脂肪组织,模型组、温肾阳颗粒各剂量组切除双侧卵巢,饲养8周后,正常组及模型组给予蒸馏水灌胃,温肾阳颗粒各剂量组给予相应剂量的温肾阳颗粒汤剂灌胃8周后取材,分别进行血清骨代谢指标检测、Micro-CT检测和病理组织学染色以及生物力学检测。结果:子宫大小称量结果显示,与模型组比较,温肾阳颗粒中剂量组小鼠子宫质量显著增加,差异有统计学意义(P<0.05)。血清骨代谢指标显示,温肾阳颗粒各剂量组体内E2水平上升,特别是中剂量组升高明显,差异有统计学意义(P<0.05)。Micro-CT结果显示,温肾阳颗粒中剂量组提高骨量最为明显;温肾阳颗粒中剂量组腰椎骨小梁体积/样本体积(BV/TV)和骨小梁厚度(Tb.Th)及骨小梁数目(Tb.N)显著高于模型组,差异有统计学意义(P<0.05),骨小梁间距(Tb.Sp)显著低于模型组,差异有统计学意义(P<0.05)。病理组织学染色结果显示,温肾阳颗粒对去卵巢小鼠的骨髓腔脂肪含量、骨小梁结构得到显著改善,骨小粱断裂的数量明显少于模型组,骨小梁密度增加,厚度变厚,间隙减小。股骨生物力学检测结果显示,与模型组比较,温肾阳中剂量组明显提高最大载荷和最大应力及弹性模量,差异有统计学意义(P<0.05)。结论:温肾阳颗粒能促进骨形成,抑制骨吸收,提高骨量和骨强度,从而起到改善骨质疏松的作用。Objective:To observe the curative efficacy of Wen-Shenyang granule on ovariectomized mice.Methods:60 female mice,3-months-old SPF,grade C57BL/6 strain,were selected.The mice were randomly divided into 5 groups:control group,model group,Wen-Shenyang granule low,middle and high doses group,with 12 in each group.The control group only resected the adipose tissue around the ovary,and the model group and Wen-Shenyang granule groups resected both ovaries.After 8 weeks of feeding,the normal group and the model group were given distilled water by gavage,and each dose group of Wen-Shenyang granule was given the corresponding dose of Wen-Shenyang granule decoction by gavage for 8 weeks.The samples were taken for serum bone metabolism index detection,Micro-CT detection,histopathological staining and biomechanical detection respectively.Results:The results of weighing uterus showed that compared with the model group,the weight of uterus in the middle dose group of Wen-Shenyang granule increased significantly(P<0.05).Serum bone metabolism index showed that the E2 level of Wen-Shenyang granule increased in each dose group,especially in the middle dose group(P<0.05).Micro-CT results showed that the middle dose group of Wen-Shenyang granule improved bone mass most obviously.The BV/TV and Tb.Th in lumbar vertebrae were significantly higher in Wen-Shenyang middle-dose group than that in model group(P<0.05),and Tb.Sp in Wen-Shenyang middle-dose group significantly lower than that in model group(P<0.05).The results of histopathological staining showed that Wen-Shenyang granules significantly improved the bone marrow cavity fat content and trabecular structure in ovariectomized mice.The number of osseointegration fractures was significantly less than that of the model group.The density of trabecular bone increased,the thickness became thicker,and the gap decreased.The results of bone biomechanics showed that compared with the model group,the maximum load,maximum stress,and elastic modulus in the middle dose Wen-Shenyang granule group
