摘 要:目的分析溃疡性结肠炎(ulcerative colitis,UC)大鼠模型中外周血、脾脏、肠道黏膜固有层淋巴细胞亚群Cx43和Cx45的表达。方法建立DNBS诱导的UC大鼠模型,ELISA检测血清IL-4和IFN-γ的表达,流式细胞术检测大鼠外周血、脾脏、肠道黏膜固有层淋巴细胞亚群Cx43和Cx45的表达。结果UC大鼠中血清IL-4水平较对照组明显降低,血清IFN-γ水平较对照组大鼠显著升高(P<0.05)。UC大鼠中,外周血淋巴细胞中CD4^(+)-Cx43、CD4^(+)-Cx45、CD8^(+)-Cx43、CD8^(+)-Cx45的表达量与对照组之间差异无统计学意义(P>0.05);UC大鼠脾脏淋巴细胞中CD4^(+)-Cx43、CD4^(+)-Cx45、CD8^(+)-Cx43、CD8^(+)-Cx45的表达量较对照组显著升高(P<0.01);UC大鼠肠道黏膜固有层淋巴细胞CD4^(+)-Cx43、CD4^(+)-Cx45表达量与较对照组显著升高(P<0.01),而CD8^(+)-Cx43、CD8^(+)-Cx45的表达量与对照组之间差异无统计学意义(P>0.05)。结论连接蛋白通过脾脏和肠道黏膜固有层淋巴细胞参与UC的免疫反应,可能作为潜在的预测标志物和预防治疗的靶点。Objective To analyze the expressions of Cx43 and Cx45 lymphocyte subsets in the lamina propria of peripheral blood,spleen and intestinal mucosa in the rat model of ulcerative colitis(UC).Methods UC rat model induced by DNBS was established,the expressions of IL-4 and IFN-γwere detected by ELISA.The expressions of Cx43 and Cx45 lymphocyte subsets in peripheral blood,spleen and the lamina propria of intestinal mucosa were detected by flow cytometry.Results In the model of UC,the serum IL-4 level was significantly lower than that in the control group,and the level serum IFN-γwas significantly higher than that of the control group(P<0.05).There was no difference in the expressions of CD4^(+)-Cx43,CD4^(+)-Cx45,CD8^(+)-Cx43 and CD8^(+)-Cx45 in peripheral blood lymphocytes between the two groups(P>0.05);The expressions of CD4^(+)-Cx43,CD4^(+)-Cx45,CD8^(+)-Cx43 and CD8^(+)-Cx45 in spleen lymphocytes of rats with UC were significantly higher than those of the control group(P<0.01).The expressions of CD4^(+)-Cx43 and CD4^(+)-Cx45 in the lamina propria of intestinal mucosa of rats with UC were significantly higher than those of the control group(P<0.01),while there were no difference in the expressions of CD8^(+)-Cx43 and CD8^(+)-Cx45 between the two groups(P>0.05).Conclusion Connexin participates in the immune response of UC through spleen and intestinal mucosal lamina propria lymphocytes,which may be a potential predictive marker and a target for prevention and treatment.
