摘 要:目的 探讨柔弱斑种草石油醚部位(PB)的抗菌和抗肺癌活性。方法 采用气相色谱质谱联用(GC-MS)法分析PB化学成分;采用微量肉汤稀释法检测PB对4种细菌活性的影响;采用MTS法筛选PB对5种肿瘤细胞增殖的影响,并通过网络药理学初步探讨其抗肺癌作用机制。结果 共鉴定出30个化学成分,主要包括亚油酸(20.13%)、十六酸(19.87%)、α-亚麻酸(13.60%)等。对大肠埃希菌、金黄色葡萄球菌金黄亚种、肠沙门氏菌肠亚种、铜绿假单胞菌活性均无明显抑制作用;对肺癌细胞A549和肝癌细胞SMMC-7721的半数抑制浓度(IC50)分别为(76.43±11.33)μg/mL和(47.20±7.67)μg/mL;网络药理学分析共筛选出肺癌疾病靶点23 727个,PB成分靶点513个,两者共有靶点504个,基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析预测到343个条目、163条信号通路,涉及配体激活的转录因子活性、神经递质受体活性,以及钙信号通路、TRP通道的炎性介质调节等通路。结论 PB主要含有脂肪酸、脂肪醇类等化学成分,无明显抗菌作用,其抗肺癌活性可能与钙信号通路、TRP通道的炎性介质调节等信号通路有关。Objective To investigate the antibacterial and anti-lung cancer activities of petroleum ether part from Bothriospermum zeylanicum(PB).Methods The chemical constituents of PB was analyzed by the gas chromatography-mass spectrometry(GC-MS).The effect of PB on the activity of four bacteria was researched by the broth microdilution method.The activity of PB on the proliferation of five tumor cells was screened by the MTS method,and its anti-lung cancer mechanism was preliminarily explored based on network pharmacology.Results A total of 30 chemical constituents were identified,including linoleic acid(20.13%),hexadecanoic acid(19.87%),α-linolenic acid(13.60%)and others.PB had no significant inhibitory effect on the activities of Escherichia coli,Staphylococcus aureus subsp.aureus,Salmonella enterica subsp.enterica and Pseudomonas aeruginosa.The half-inhibitory concentration(IC50)of PB on the lung cancer A549 cells and liver cancer SMMC-7721 cells were(76.43±11.33)μg/mL,(47.20±7.67)μg/mL respectively.The network pharmacology analysis showed that 23727 lung cancer targets,513 constituent targets of PB and 504 common targets were screened;there were 343 items and 163 signaling pathways were obtained through the gene ontology(GO)functional enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis,involving ligand-activated transcription factor activity,neurotransmitter receptor activity,calcium signaling pathways and inflammatory mediator regulation of TRP channels.Conclusion PB mainly contains chemical constituents such as fatty acids and fatty alcohols,and has no significant antibacterial effect.Its anti-lung cancer activity may be related to the calcium signaling pathways and inflammatory mediator regulation of TRP channels.
