摘 要:目的:分析不同细胞系对胃癌细胞外泌体的摄取差异。方法:使用荧光蛋白融合hCD63分子标记外泌体,并筛选标记的稳定细胞株。使用超速离心收集外泌体,电镜观察荧光蛋白标记外泌体外形。标记的外泌体与不同细胞系细胞孵育后,使用流式细胞术分析细胞携带荧光的差异。结果:通过流式细胞分析证明荧光蛋白在hCD63分子的羧基端进行标记,可以获得最大的荧光强度。不同细胞系对MGC-803外泌体的摄取存在差异。结论:单核细胞样的THP-1细胞摄取胃癌细胞外泌体的能力强于其他组织来源的细胞系。Objective:To analyze the differences in the uptake of gastric cancer-derived exosomes by different cell lines.Methods:Fluorescent fusion hCD63 was used to label exosomes and screen labeled stable cell line.The labeled exosomes were prepared by ultracentrifugation.A transmission electron microscope was used to further observe the morphology of fluorescent-proteinlabeled exosomes.The exosomes were co-incubated with different cell lines.Flow cytometry was hired to analyze the difference in fluorescence signal of different cell lines.Results:Flow cytometry proved that the fluorescent protein fused to hCD63 at the carboxyl terminus can emit the strongest fluorescent signal.The uptake of gastric cancer-derived exosomes by of different cell lines was different.Conclusion:The ability of monocyte-like cell line THP-1 to uptake gastric cancer-derived exosomes is greater than that of other cell lines.
