摘 要:目的了解本地区副溶血性弧菌的毒力基因和优势血清型,为防控副溶血性弧菌引起的感染性腹泻提供分子学依据。方法去收集2019年-2022年腹泻患者、食品安全风险监测样品中分离得到的共计145株副溶血性弧菌,根据GB4789.7—2013《食品安全国家标准食品微生物学检验副溶血性弧菌检验》进行0抗原和K抗原血清分型;采用荧光定量PCR检测耐热直接溶血毒素基因(tdh)、不耐热溶血素基因(tlh)、耐热直接溶血素相关基因(trh)。结果145株副溶血性弧菌中,123株分型明确,其中以03:K6最多,占60.00%,04:K8其次,占22.75%。tlh、tdh、trh基因的检出率分别为100.00%、75.17%、2.76%;03:K6型和04:K8型毒力基因检出率较高,两者差异无统计学意义(P>0.05)。03:K6和04:K8型总体毒力基因检出率为86.67%,高于其他血清型(24.00%),差异有统计学意义(P<0.05)。病例分离株的毒力基因检出率(100.00%)高于食品分离株(5.41%)(Х^(2)=134.67,P<0.05)。结论本地区流行的副溶血性弧菌优势血清型是03:K6和04:K8,血清型与毒力基因检出率有相关性;病例分离株毒力基因检出率高于食品分离株。Objective This paper aims to understand the virulence gene and dominant serotype of Vibrio parahaemolyticus in this region,so as to provide molecular basis for the prevention and control of infectious diarrhea caused by Vibrio parahaemolyticus.Methods A total of 145 Vibrio parahaemolyticus isolated from diarrhea patients and food safety risk monitoring samples were collected from 2019 to 2022.According to GB 4789.7-2013 National Food Safety Standard:Microbiological Examination of Vibrio parahaemolyticus,serotyping of O antigen and K antigen was performed.Fluorescent quantitative PCR was used to detect thermostable direct hemolysin gene(tdh),thermostable direct hemolysin gene(tlh)and thermostable direct hemolysin related gene(trh).Results In 145 strains of Vibrio parahaemolyticus,123 strains were clearly classified,03:K6 accounted for 60.0%,followed by 04:K8,accounting for 22.75%.The detection rates of tlh,tdh and trh genes were 100.00%,75.17%and 2.76%respectively.The detection rate of virulence genes of 03:K6 and 04:K8 is high,there was no statistical significance on the difference between them(P>0.05).The overall detection rate of virulence genes of 03:K6 and 04:K8 was 86.67%,much higher than that of other serotypes(24.00%)(P<0.05).The detection rate of virulence gene was 100.00%,which was much higher than that of food strain(5.41%)(Х^(2)=134.67,P<0.05).Conclusion The dominant serotypes of Vibrio parahaemolyticus prevalent in this region are 03:K6 and 04:K8.The serotypes are correlated with the detection rate of virulence gene,and the detection rates of virulence genes from case strains and food strains were different.
