摘　　要：目的 探讨METTL3在胃癌中的表达及意义。方法 收集Oncomine数据库中关于METTL3的数据信息,对其在胃癌中的作用进行统计分析。利用Kaplan-Meier Plotter进行患者生存周期分析。结果 经过筛选,涉及mRNA METTL3在正常胃组织和胃癌组织表达差异的研究共4项(其中按照病理分为9项,共包含346例组织样本),与对照组相比,METTL3在346例胃癌组织中高表达(P<0.05)。METTL3表达量与胃癌总体生存呈负相关,METTL3表达水平越高则患者总体生存率越低(P<0.05)。根据胃癌Lauren分型探讨,发现肠型胃癌以及混合型胃癌中METTL3的表达情况与生存时间之间关系不大(P>0.05)。弥漫型胃癌中,METTL3的表达水平与预后具有相关性,且METTL3表达水平高的患者生存更获益(P<0.05)。针对HER2阳性的胃癌,METTL3的表达水平与胃癌患者预后呈负相关(P<0.05)。结论 mRNA METTL3在胃癌组织中高表达,且与胃癌预后有关,可能为肿瘤药物的开发提供重要理论依据,但需要更多的研究进行探讨。Objective To investigate the expression and significance of METTL3 in gastric cancer.Methods The data of METTL3 in Oncomine database was collected and the effect of METTL3 in gastric cancer was analyzed statistically.Kaplan-Meier Plotter was used to analyze the survival cycle of patients.Results After seriously screening,a total of 4 studies involving the difference of mRNA METTL3 expression in normal stomach tissues and gastric cancer tissues(according to pathology there were 9 studies,including 346 tissue samples)were conducted.Compared with the control group,METTL3 was highly expressed in 346 gastric cancer tissues(P<0.05).METTL3 expression level was negatively correlated with overall survival of gastric cancer,and the higher the METTL3 expression level,the lower the overall survival rate of patients(P<0.05).According to Lauren's classification of gastric cancer,it was found that there was little relationship between METTL3 expression and survival time in intestinal and mixed gastric cancer(P>0.05).In diffuse gastric cancer,METTL3 expression level ass correlated with prognosis,and patients with high METTL3 expression level benefit more from survival(P<0.05).For HER2-positive gastric cancer,the expression level of METTL3 was negatively correlated with the prognosis of patients with gastric cancer(P<0.05).Conclusions mRNA of METTL3 is highly expressed in gastric cancer tissues,and is associated with the prognosis of gastric cancer,which may provide important theoretical basis for the development of anticancer drugs,but more studies are needed for the investigation.
