美洲大蠊提取液对兔角膜上皮损伤修复影响的实验研究

摘　　要：目的探讨美洲大蠊提取液(PAE)对兔角膜上皮损伤修复的影响。方法乙醇法建立新西兰大白兔角膜上皮损伤模型。随机分为0h组、PAE组、人表皮生长因子(hEGF)滴眼液组和生理盐水(NS)组。使用Image-J软件测量荧光染色面积,病理切片观察各组不同时间点角膜上皮修复情况。结果造模后72h,PAE组与hEGF组的角膜上皮修复速率与NS组相比均更快(P=0.001、0.034)。造模后36、48h,PAE组与hEGF滴眼液组角膜上皮愈合率高于NS组(P=0.005、0.028)。病理切片结果表明,造模后72h,相较于其它两组,PAE组角膜上皮层更厚,细胞之间排列更加紧密。结论PAE能促进兔角膜上皮损伤的修复,能显著缩短角膜上皮愈合时间,促进角膜上皮重塑。Objective To investigate the effects of Periplaneta Americana extract(PAE)on the repair of corneal epithelium injury in rabbits.Methods The corneal epithelium injury model of New Zealand white rabbits was established by the ethanol method,and divided into a PAE group,a human epidermal growth factor(hEGF)eye drops group and normal saline(NS)group randomly.The staining area with fluorescein sodium was measured by Image-J software.Corneal histopathological sections were prepared to compare the repair of corneal epithelium at different time points in each group.Results The rate of corneal epithelial repair in the PAE group and hEGF eye drops group was faster than that in the NS group within 72 hours(P=0.001,0.034).The healing rate of corneal epithelium of the PAE group and hEGF eye drops group was higher than that in the NS group at 36 and 48 hours(P=0.005,0.028).The results of histopathological examination showed that the corneal epithelium in PAE group was thicker and the epithelium cells were more closely arranged 72h after modeling compared with hEGF eye drops group and NS group.Conclusion PAE can promote the repair of corneal epithelium injury in rabbits,significantly shorten complete healing of corneal epithelium,and promote the corneal epithelium remodeling after the corneal epithelium injury.
