散发性结直肠癌患者血清LncRNA GACAT3 miR-485-5p表达及其临床意义

摘　　要：目的探讨散发性结直肠癌患者血清长链非编码RNA GACAT3(LncRNA GACAT3)、微小RNA-485-5p(miR-485-5p)的表达及其临床意义。方法对62例散发性结直肠癌患者(研究组)、53名健康志愿者(对照组)采用实时荧光定量聚合酶链反应检测GACAT3、miR-485-5p的表达;采用LncBase网站对GACAT3与miR-485-5p的结合进行预测,并采用Pearson相关分析法分析GACAT3与miR-485-5p的相关性;根据GACAT3与miR-485-5p相对表达量平均值将研究组患者分为GACAT3高表达组(29例)、GACAT3低表达组(33例)和miR-485-5p高表达组(30例)、miR-485-5p低表达组(32例),分析不同临床特征患者GACAT3、miR-485-5p表达情况;采用受试者工作特征(ROC)曲线分析血清GACAT3、miR-485-5p对散发性结直肠癌的诊断价值;采用Kaplan-Meier曲线分析散发性结直肠癌患者5 a生存情况。结果研究组血清GACAT3相对表达量高于对照组,血清miR-485-5p相对表达量低于对照组(P<0.01)。散发性结直肠癌患者GACAT3与miR-485-5p存在互补结合的核苷酸位点,GACAT3与miR-485-5p呈显著负相关(r=-0.306,P<0.05)。肿瘤不同分化程度、淋巴结转移情况、TNM分期患者血清GACAT3、miR-485-5p表达情况比较,差异有统计学意义(P<0.05或0.01)。ROC曲线显示,血清GACAT3与miR-485-5p联合诊断散发性结直肠癌的曲线下面积为0.918,95%CI为0.865~0.970,诊断敏感度为0.806,诊断特异度为0.887。GACAT3低表达组平均生存期高于高表达组,miR-485-5p低表达组平均生存期低于高表达组。结论散发性结直肠癌患者血清GACAT3表达上调,miR-485-5p表达下调,二者呈负相关,并可能作为散发性结直肠癌诊断及评估患者预后的重要指标。Objective To explore the expressions of serum LncRNA GACAT3 and miR-485-5p in patients with sporadic colorectal cancer(CRC)and clinical significance.Methods The expression levels of GACAT3 and miR-485-5p were detected in 62 CRC patients(research group)and 53 healthy volunteers(control group)using quantitative real-time PCR(qRT-PCR);LncBase website was used to predict the combination of GACAT3 and miR-485-5p;the correlation between GACAT3 and miR-485-5p was analyzed by Pearson method.According to the averages of relative expression levels of GACAT3 and miR-485-5p patients were grouped into high GACAT3(n=29),low GACAT3(n=33),high miR-485-5p(n=30)and low miR-485-5p(n=32)expression,their correlations with clinical pathological parameters analyzed;diagnostic values of serum GACAT3 and miR-485-5p in sporadic CRC were analyzed using the receiver operating characteristic(ROC)curve;5-year survival situations of the CRC patients were analyzed adopting Kaplan-Meier curve.Results The relative expression level of serum GACAT3 was higher in experimental than control group and difference statistically significant(P<0.01);the relative expression level of miR-485-5p was lower in experimental than control group and difference statistically significant(P<0.01).There were complementary binding nucleotide sites between GACAT3 and miR-485-5p,GACAT3 was significantly negatively related to miR-485-5p(r=-0.306,P<0.05);the expressions of serum GACAT3 and miR-485-5p were related to differentiated degree,lymph node metastasis,and TNM staging(P<0.05 or 0.01).Area under the ROC of combined diagnosis of serum GACAT3 and miR-485-5p in the CRC was 0.918,the 95%confidence interval 0.865~0.970,diagnostic sensitivity 0.806,and diagnostic specificity 0.887.The mean survival time was higher in low than high GACAT3 expression group and that lower in low than high miR-485-5p expression group.Conclusion The expression of serum GACAT3 of sporadic CRC patient up-regulates and that of miR-485-5p down-regulates,the two are negatively correlated,and may be u
