摘 要:为了筛选出潜在的肝癌检测标志物,从基因表达数据库(Gene Expression Omnibus database,GEO)中选择GSE14520、GSE36376、GSE25097数据集,筛选出公共差异表达基因,利用DAVID平台进行富集分析,应用Cytoscape软件筛选出枢纽基因,并通过GEPIA2平台对枢纽基因的表达和预后进行验证,最后分析蛋白表达水平.共197个基因被定义为3个基因芯片的公共差异表达基因,其中,CDC20、RACGAP1、ASPM、RFC4、FEN1、AURKA、NUSAP1、TOP2A为枢纽基因,均在肝癌中高表达并影响不良预后.枢纽基因的KEGG重分析显示,RFC4和FEN1显著富集在DNA复制通路;蛋白表达分析显示,肝癌患者的RFC4和FEN1基因的转录与翻译均表现为高表达.筛选出的8个基因可以作为肝癌诊断潜在的生物标志物,其中RFC4和FEN1可能是调控肝癌发生、发展与转移的关键基因.In order to screen potential biomarkers for liver cancer detection,the GSE14520,GSE36376,and GSE25097 datasets were selected from the Gene Expression Omnibus database to screen common differentially expressed genes.The DAVID platform was used for enrichment analysis,and the Cytoscape software was used to screen the hub genes.The expression and prognosis of the hub genes were validated using the GEPIA2 platform,and finally the protein expression level was analyzed.A total of 197 genes were defined as common differentially expressed genes in three gene chips,among which CDC20,RACGAP1,ASPM,RFC4,FEN1,AURKA,NUSAP1,TOP2A were hub genes,all of which were highly expressed in liver cancer and affected poor prognosis.KEGG reanalysis of hub genes showed that RFC4 and FEN1 were significantly enriched in DNA replication pathway,and protein expression analysis showed that the transcription and translation of RFC4 and FEN1 genes in patients with liver cancer might be highly expressed.These eight genes can serve as potential biomarkers for liver cancer diagnosis,among which RFC4 and FEN1 may be key genes regulating the occurrence,development,and metastasis of liver cancer.
