摘 要:以德州驴为研究对象,利用转录组学(RNA-seq)技术对其背最长肌、腿后腱、臀大肌不同部位差异表达基因进行测序,筛选调控驴肉品质性状的关键基因,探究不同部位驴肉差异的分子机制。结果表明:筛选获得了臀大肌与背最长肌差异表达基因124个,其中上调基因73个,下调基因51个;腿后腱与背最长肌差异表达基因126个,其中上调基因68个,下调基因58个。对德州驴腿后腱与背最长肌差异基因KEGG通路富集分析发现,差异表达基因主要在赖氨酸降解、β-丙氨酸代谢、脂肪酸降解等通路中高度富集,臀大肌与背最长肌差异表达基因富集氧化磷酸化、亮氨酸和异亮氨酸降解等通路中。同时,肉品质相关候选基因GCDH、ALDH2、IGF1、VGLL2、TECRL、ASS1、ATP5A1、MUT、ACADSB参与肌肉氨基酸脂肪酸代谢过程,在肌肉生长过程中发挥着重要作用。Using Dezhou donkey as research objects, the transcriptome of donkey muscle in different parts was analyzed in order to obtain the key genes related to regulating meat quality traits and provide a theoretical basis for improving donkey meat quality. The results showed that there were 124 differential genes between gluteus and longissimus dorsi muscle, including 73 up-regulated genes and 51 down-regulated genes. There were 126 differential genes between leg muscle and longissimus dorsi muscle, including 68 up-regulated genes and 58 down-regulated genes. KEGG pathway enrichment analysis of the differential gene between Dezhou donkey leg muscle and longissimus dorsi muscle showed that the differentially expressed genes were highly enriched in lysine degradation, alanine metabolism, fatty acid degradation and other pathways, and the differentially expressed genes between gluteus muscle and longissimus dorsi muscle were enriched in oxidative phosphorylation,leucine and isoleucine degradation and other pathways.At the same time,the candidate genes GCDH,ALDH2,IGFI,VGLL2,TECRL,ASS1,ATP5A1,MUTand ACADSBrelated to meat quality were screened,which were involved in the metabolism of amino acids and fatty acids in muscle and played an important role in the process of muscle growth.
