摘  要:缺血再灌注损伤(IRI)是指血供中断再通后,组织器官损伤加重的过程,其损伤机制复杂,主要包括氧化应激、钙超载、炎性反应等,由此引起的临床疾病称为再灌注综合征。乙醛脱氢酶2(ALDH2)是人体乙醇代谢过程中的关键酶,主要功能是将乙醛代谢成无毒的乙酸。近年来有大量研究表明,ALDH2在缺血再灌注损伤中有明确的保护性作用,其特异性激动剂Alda-1也被广泛运用于IRI相关研究中。本文就ALDH2的结构、功能、基因多态性和在各器官IRI中的研究进展进行了综述,探讨ALDH2成为IRI治疗靶点的可能性。Ischemia reperfusion injury(IRI)refers to the process of tissue and organ damage aggravated after blood supply is interrupted and rerouted.The injury mechanism is complex,including oxidative stress,calcium overload,inflammatory response,etc.,and the clinical diseases caused by IRI are called reperfusion syndrome.Acetaldehyde dehydrogenase 2(ALDH2)is a key enzyme in the process of ethanol metabolism,and its main function is to metabolize acetaldehyde into non-toxic acetic acid.In recent years,a large number of studies have shown that ALDH2 has a clear protective effect on ischemia-reperfusion injury,and its specific agonist Alda-1 has also been widely used in IRI related studies.In this paper,the structure,function,gene polymorphism of ALDH2 and the research progress in IRI were reviewed,and the possibility of ALDH2 as a therapeutic target of IRI was discussed.
