摘  要:目的:建立同时检测双孢蘑菇中4种嘌呤含量的高效液相色谱法。方法:取0.25 g冷冻干燥后的双孢蘑菇粉,加入5%的乙醇溶液25 mL超声提取30 min,冷却至室温,用无水乙醇定容至25 mL,用中性滤纸过滤后再经0.45μm膜过滤去除沉淀,滤液上机测定,分析双孢蘑菇中4种嘌呤含量。结果:4种嘌呤能够完全分离,且在质量浓度0.1~20.0 mg·L^(-1)时,4种嘌呤的响应峰面积与其浓度呈线性关系,r^(2)> 0.995,相对标准偏差<3%,回收率98.86%~103.25%。结论:该分析方法简便、准确,可同时检测双孢蘑菇中4种嘌呤的含量。Objective:To establish a high-performance liquid chromatography method for the simultaneous detection of four purines in Agaricus bisporus.Method:Take 0.25 g of freeze-dried Agaricus bisporus powder,add 25 mL of 5%ethanol solution and extract it by ultrasound for 30 min.Cool to room temperature,dilute to 25 mL with anhydrous ethanol,filter with neutral filter paper,and then pass through 0.45μm membrane filtration,removal of sediment,determination of the filtrate on the machine,and analysis of the content of four purines in Agaricus bisporus.Result:Four purines can be completely separated,and in the mass concentration range of 0.1~20.0 mg·L^(-1),the response peak area of four purines is linear with their concentration,r^(2)>0.995,precision RSD<3%,recovery rate 98.86%~103.25%.Conclusion:The method is simple and accurate,and can be used to simultaneously detect the contents of four purines in Agaricus bisporus.
