摘 要:脂肪肝病是一种营养代谢疾病,根据病因划分为酒精性脂肪肝病(alcoholic fatty liver disease,AFLD)和非酒精性脂肪肝病(non-alcoholic fatty liver disease,NAFLD)。为了比较小鼠AFLD与NAFLD模型的损伤差异,本研究利用昆明系小鼠分别进行造模。其中AFLD组小鼠饲喂60%高脂饲料,结合每天灌胃56%浓度及50%浓度白酒1次,连续灌胃35 d;NAFLD组小鼠饲喂60%高脂饲料,持续饲喂12周。造模期结束后观察小鼠肝脏损伤状况,检测生化相关指标及脂代谢相关基因变化情况。结果显示:AFLD组小鼠在造模前期死亡率很高,体质量增长缓慢,NAFLD组小鼠体质量显著增长;AFLD组和NAFLD组血清肝功能指标明显增长,AFLD组血清血脂指标下降,NAFLD组血清血脂指标升高;NAFLD组小鼠肝脏组织发生脂肪变性;AFLD组、NAFLD组肝脏脂代谢相关基因PPAR-γ和NR1H4表达均下调,AFLD组肝脏炎症相关基因TLR4表达下调,NAFLD组TLR4表达上调。本研究为探索乙醇和高脂造成肝脏损伤的机制提供新的思路。Fatty liver disease is a nutritional metabolic disorder that is classified according to the causes into Alcoholic fatty liver disease(AFLD)and Non-alcoholic fatty liver disease(NAFLD).In order to compare the damage differences between the AFLD and NAFLD models,the study used Kunming mice to mold separately.Mice in the AFLD group were fed 60% high-fat feed,and were gavaged with 56% and 50% liquor per day for 35 days,while mice in the NAFLD group were fed 60% high-fat feed for 12 weeks.After the end of the molding period,the liver damage of mice was observed,and the biochemical-related indicators and changes in lipid metabolism-related genes were detected.According to the results,the mice in the AFLD group had a high mortality rate in the early stage of the experiment and the body mass increased slowly,while the body mass of the mice in the NAFLD group increased significantly.Serum hepatic function indexes in both the AFLD and NAFLD groups increased significantly;serum lipid index decreased in the AFLD group while that in the NAFLD group increased.Hepatic steatosis occurred in NAFLD group.The expression of liver lipid metabolism-related genes PPAR-γ and NR1H4 in both the AFLD and NAFLD groups were down-regulated;the liver inflammation-related gene TLR4 expression in the AFLD group was down-regulated while that in the NAFLD group was up-regulated.This study provides new ideas for discovering the mechanism of liver injury caused by ethanol and high fat.
