肝郁脾虚因素刺激对DEN诱发肝癌大鼠免疫功能的影响

摘　　要：观察肝郁脾虚因素刺激对二乙基亚硝胺 (DEN)诱发大鼠实验性肝癌的发生及T淋巴细胞功能的影响 ,以及用疏肝健脾方药对该模型的防治作用。大鼠经肝郁脾虚因素刺激和DEN 70mg/kg灌胃 ,每周 1次 ,连续 8周后停用DEN ,预防和治疗组给予疏肝健脾方药 2mL/只灌胃 (1 5g/kg)。大鼠分别在实验第 9、14、19周取脾测定T淋巴细胞增殖和产生IFN γ和IL 2的能力。结果显示 :大鼠经肝郁脾虚因素刺激和DEN作用下 ,与单纯DEN诱发肝癌大鼠比较 ,T淋巴细胞增殖和产生IFN γ和IL 2能力的显著降低 (P值分别 <0 0 5 ) ;然而 ,经疏肝健脾方药防治后大鼠T淋巴细胞增殖能力和产生IL 2、IFN γ的能力恢复明显。说明应用疏肝健脾方药能显著改善肝郁脾虚因素刺激对DEN诱发肝癌大鼠的免疫功能。The influence of the factors of liver depression and spleen deficiency (LDSD) on the occurrence of hepatocarcinoma induced by diethylnitrosamine (DEN) and on the function of T lymphocytes, and the treating and preventing effects of the Shuganjianpi Formula (SF) were investigated in the model rats. All rats in the experiment were stimulated with the factors of LDSD for 19 weeks, and simultaneously, given by gastric perfusion a dose of DEN, 70 mg/kg, once a week for 8 weeks; the rats in the preventing and treating group were given a dose of SF, 1 5 g/ kg; and then, the abilities of T lymphocytic proliferation and IFN γ and IL 2 production were detected in the isolated spleen of the rats in the 9th, 14th and 19th weeks respectively. The results showed that the abilities of T lymphocytic proliferation and IFN γ and IL 2 production were significantly lower in the rats stimulated with both the factors of LDSD and DEN than those in the rats stimulated only with DEN ( P <0 05); but after the rats were given SF, the abilities of T lymphocytic proliferation and IFN γ and IL 2 production were significantly restored. The results suggest that SF can significantly improve immunological function in the rats with DEN induced hepatocarcinoma plus the stimulation of the LDSD factors, and at the same time, can relieve the severity of hepatocarcinoma.
