摘 要:目的:筛选结直肠癌相关的lncRNA,构建lncRNA-miRNA-mRNA竞争性内源网络,并预测其生物学功能。方法:从GEO数据库下载结直肠癌表达谱芯片数据并进行差异分析,使用Cytoscape软件构建ceRNA调控网络,利用DAVID对差异基因进行基因本体论(GO)功能和京都基因和基因组百科全书(KEGG)通路富集分析,使用GEPIA数据库验证差异表达lncRNA与结直肠癌患者预后的关系。结果:从GEO数据库下载芯片GSE156720、GSE126092、GSE156732和GSE134834,筛选出差异表达基因,构建了基于4个lncRNA(HAND2-AS1、MEF2C-AS1、LINC01354和CRNDE)的ceRNA调控网络。GO、KEGG以及蛋白质互作分析结果显示这4个lncRNA可能通过竞争性结合miRNA,参与到结直肠癌的发生发展过程,其中HAND2-AS1与结直肠癌患者预后有关(P<0.05)。结论:通过生物信息学方法构建了结直肠癌lncRNA相关的ceRNA调控网络,为研究结直肠癌的分子机制研究提供了参考。Objective:To screen lncRNAs associated with colorectal cancer,construct competitive endogenous networks of lncRNA-miRNA-mRNA,and predict the biological functions.Methods:Colorectal cancer expression profile chip data was downloaded from GEO database and differential expression analysis was performed.Cytoscape software was used to construct ceRNA regulatory network.DAVID was used to perform gene ontology(GO)function and KEGG pathway enrichment analysis of differentially expressed genes.GEPIA database was used to verify the relationship between differentially expressed lncRNAs and prognosis of colorectal cancer patients.Results:GSE156720,GSE126092,GSE156732 and GSE134834 were downloaded from GEO database.Deferentially expressed genes were screened out,and a ceRNA regulatory network based on four lncRNAs(HAND2-AS1,MEF2C-AS1,LINC01354 and CRNDE)was constructed.The results of GO,KEGG and protein interaction analysis showed that these four lncRNAs might associated with the occurrence and development of colorectal cancer through competitive binding of miRNA.HAND2-AS1 was associated with the prognosis of colorectal cancer patients(P<0.05).Conclusion:The lncRNA-related ceRNA regulatory network of colorectal cancer is constructed by bioinformatics methods,which provide references for further study of the molecular mechanism of colorectal cancer.
