摘 要:目的:为探究绣球菌冻干片对S180荷瘤小鼠的抗肿瘤作用及机理。方法:选择破壁灵芝孢子粉和茯苓多糖口服液作为阳性对照,将50只S180荷瘤小鼠随机分为模型组(Model)、破壁灵芝孢子粉组(Ganoderma lucidum Spore Powder,GS)、茯苓多糖组(Poria cocos Polysaccharides,PCP)、绣球菌低剂量组[0.31 g/(kg·d),Sparassis crispa Low-Dose Group,SCL]和绣球菌高剂量组[0.62 g/(kg·d),Sparassis crispa High-Dose Group,SCH],连续灌胃样品18 d,测定小鼠体重、肿瘤重量和体积,检测脏器指数、白细胞计数、细胞因子(IL-2、IL-6、IL-8和TNF-α)浓度、脾淋巴细胞增殖活性、自然杀伤(Natural killer,NK)细胞活性和结肠内容物中短链脂肪酸(Short Chain Fatty Acids,SCFAs)水平,并分析瘤体指数与免疫功能指标的相关性。结果:GS、PCP、SCL和SCH的肿瘤抑制率分别为52.2%、36.7%、37.4%和49.0%,3种试验样品均可提高荷瘤小鼠胸腺指数、促进IL-2和TNF-α分泌、增强脾淋巴细胞增殖活性和NK细胞杀伤活性,且绣球菌冻干片高剂量优于低剂量,但3种样品对荷瘤小鼠结肠内SCFAs浓度无明显影响。Pearson相关性分析发现瘤体指数与IL-2、TNF-α水平负相关(P<0.05)。结论:绣球菌冻干片抑制S180肿瘤作用可能是通过非SCFAs依赖途径的增强小鼠细胞免疫和体液免疫功能实现。Objective:To explore the anti-tumor effects of freeze-dried tablets containing Sparassis crispa(FSC)on S180-bearing mice and reveal the underlying mechanism.Method:Broken-Ganoderma lucidum spore powder(GS)and Poria cocos polysaccharides(PCP)were selected as positive anti-tumor samples.Fifty S180 tumor-bearing mice were randomly divided into five groups:model group(Model),GS group,PCP group,FSC low-dose group[0.31 g/(kg·d),SCL],and FSC high-dose group[0.62 g/(kg·d),SCH],and were orally administrated daily with testing samples for 18 consecutive days.Subsequently,the body weight,the weight and volume of the tumor,the organ index,the white blood cell count,the cytokine(IL-2,IL-6,IL-8,and TNF-α)concentrations,the spleen lymphocyte proliferative activity,the natural killer(NK)cell activity,and the level of short chain fatty acids(SCFAs)in colon content of S180-bearing mice were measured.Finally,the correlation between tumor index of mice and immune function indicators was analyzed.Result:The tumor inhibition rate of GS,PCP,SCL,and SCH groups was 52.2%,36.7%,37.4%,and 49.0%,respectively.The results demonstrated that the three testing samples could increase the thymus index,promote IL-2 and TNF-αsecretion,enhance the proliferation activity of splenic lymphocytes,and improve splenic NK cell activity.Particularly,the improvements in immune functionsof S180-bearing in the SCH group were better than those in the SCL group.However,there were no significant differencesin the concentrations of colonic SCFAs among the three testing samples.Pearson analysis revealed that the degrees of tumorindex were negatively correlated with the concentrations of IL-2 and TNF-α(P<0.05).Conclusion:The anti-tumor effectsof FSC might be achieved by enhanced cellular and humoral immune functions of S180-bearing mice via non-SCFAsdependentmechanisms.
