天麻对APP/PS1痴呆小鼠模型的神经保护作用及其对衰老相关蛋白的影响

摘  要:目的探讨天麻(RG)对APP/PS1双转基因小鼠脑组织衰老相关蛋白水平的影响及其神经保护作用。方法30只C57正常小鼠随机分为正常组、正常给药组;30只APP/PS1模型小鼠随机分为模型组、模型给药组;正常给药组、模型给药组灌胃给予1.5 g/(kg·d)的RG,各组小鼠6月龄时进行水迷宫实验;免疫组织化学检测脑组织内Aβ聚集;Western印迹测定脑组织中衰老相关蛋白p53、p21表达水平;酶联免疫吸附试验(ELISA)检测各组脑组织中白细胞介素(IL)-6、IL-8表达。结果与正常组比较,模型组学习记忆能力显著降低,脑组织内Aβ斑块聚集明显增多,p21、IL-6、IL-8表达显著增加(P<0.05,P<0.01)。与模型组比较,模型加药组学习记忆能力显著升高,脑组织内Aβ斑块聚集明显减少,p21、IL-6、IL-8表达显著降低(P<0.05,P<0.01)。结论RG可能通过调控衰老相关蛋白及IL-6、IL-8表达延缓衰老进程,改善阿尔茨海默病(AD)病理进程。Objective To explore the effect of rhizome gastrodiae(RG)on the levels of aging-related proteins in the brain tissue of APP/PS1 double transgenic mice and its neuroprotective effect.Methods Thirty C57 normal mice were randomly divided into normal group and normal dosing group;thirty APP/PS1 model mice were randomly divided into model group and model dosing group,normal dosing group and model dosing group were given 1.5 g/(kg·d)RG by gavage,mice in each group were subjected to a water maze experiment at the age of 6 months;immunohistochemistry was used to detect Aβaggregation in the brain tissue;Western blotting was used to determine the aging-related protein p53,p21 expression levels in brain tissue;enzyme linked immunosorbent assay(ELISA)was used to detect the expressions of interleukin(IL)-6 and IL-8 in the brain tissue of each group.Results Compared with the normal group,the learning and memory ability of the model group were significantly reduced,the Aβplaque accumulation in the brain tissue was significantly increased,and the expressions of p21,IL-6 and IL-8 were significantly increased(P<0.05,P<0.01);compared with the model group,the learning and memory ability were significantly increased,the Aβplaque accumulation in the brain tissue was significantly reduced,and the expressions of p21,IL-6,and IL-8 were significantly decreased in the model dosing group(P<0.05,P<0.01).Conclusions RG might delay the aging process and improve the pathological process of Alzheimer′s disease(AD)by regulating the expression of aging-related proteins and IL-6 and IL-8.
