摘　　要：[目的]探讨KIAA1456通过靶向Notch调控对非小细胞肺癌生长、侵袭的影响。[方法]设置H1299细胞组、KIAA1456 mimics组、KIAA1456 inhibitor组,各组细胞培养48 h;培养结束后,CCK-8测量细胞增殖,流式细胞仪测定细胞凋亡水平,Transwell实验测定细胞侵袭水平,RT-PCR法及蛋白印迹法测定细胞KIAA1456、Notch1 mRNA与蛋白水平。[结果]KIAA1456 mimics组OD值、存活率、单克隆形成数目、穿膜数低于H1299细胞组,KIAA1456 inhibitor组OD值、存活率、单克隆形成数目、穿膜数高于H1299细胞组、KIAA1456 mimics组。KIAA1456 mimics组凋亡率、KIAA1456 mRNA和蛋白高于H1299细胞组(P<0.05),KIAA1456 inhibitor组凋亡率、KIAA1456 mRNA和蛋白低于H1299细胞组、KIAA1456 mimics组(P<0.05)。KIAA1456 mimics组高于H1299细胞组(P<0.05),KIAA1456 mimics组Notch1 mRNA和蛋白低于H1299细胞组(P<0.05),KIAA1456 inhibitor组Notch1 mRNA和蛋白高于H1299细胞组、KIAA1456 mimics组(P<0.05)。[结论]KIAA1456能抑制非小细胞肺癌H1299细胞增殖侵袭,促进其凋亡;其机制可能与KIAA1456抑制Notch通路的激活有关。[Objective]To investigate the effect of KIAA1456 on the growth and invasion of non-small cell lung cancer by targeting Notch.[Method]H1299 cell group,KIAA1456 mimics group,and KIAA1456 inhibitor group were set up.Cells in each group were cultured for 48 hours;after the culture,the cell proliferation was measured by CCK-8,and the cell apoptosis was measured by flow cytometry The level of cell invasion was determined by Matrigel chamber,and the mRNA and protein levels of KIAA1456 and Notch1 were determined by RT-PCR and Western Blotting.[Result]The OD,survival rate,number of monoclonal formation,and number of membrane penetrations in the KIAA1456 mimics group were lower than those in the H1299 cell group,the OD,survival rate,number of monoclonal formation,and number of membrane penetration were higher in the KIAA1456 inhibitor group than in the H1299 cell group and the KIAA1456 mimics group.Apoptosis rate,KIAA1456 mRNA and protein were higher in the KIAA1456 mimics group than in the H1299 cell group(P<0.05),and the apoptosis rate,KIAA1456 mRNA and protein were lower in the KIAA1456 inhibitor group than in the H1299 cell group and the KIAA1456 mimics group(P<0.05).mRNA and protein were higher in the KIAA1456 mimics group than in the H1299 cell group(P<0.05),Notch1 mRNA and protein were lower in KIAA1456 mimics group than in H1299 cell group(P<0.05),and Notch1 mRNA and protein were higher in KIAA1456 inhibitor group than in H1299 cell group,KIAA1456 mimics group(P<0.05).[Conclusion]KIAA1456 can inhibit the proliferation and invasion of non-small cell lung cancer H1299 cells and promote their apoptosis;the mechanism may be related to the inhibition of the activation of Notch pathway by KIAA1456.
