摘  要:[目的]研究薯蓣皂苷对肺癌细胞的生物学过程的影响及其相关机制。[方法]MTT试验及流式细胞术检测不同浓度薯蓣皂苷对A549细胞增殖及凋亡的影响,并筛选半数抑制浓度(IC50)。酶活性试剂盒技术检测细胞己糖激酶(HK)、丙酮酸激酶(PK)活性,WB检测Hippo-YAP通路蛋白表达水平。[结果]MTT的结果发现,薯蓣皂苷在24 h对A549细胞的IC50是0.08μmol/L。随着薯蓣皂苷浓度的增大,A549细胞凋亡率越高。与对照组相比,薯蓣皂苷或Veteporfin处理组的A549细胞葡萄糖摄取能力降低,乳酸及ATP产量降低(P均<0.05)。与对照组相比,薯蓣皂苷或Veteporfin处理可降低HK及PK活性(P均<0.05),并降低A549细胞内MOB1及p-YAP蛋白表达量(P均<0.05)。[结论]薯蓣皂苷可能是通过干预Hippo-YAP信号通路抑制糖酵解作用进而促进细胞凋亡。[Objective]To study the effect of diosgeninon the cellular biological processes of lung cancer cells and related mechanisms.[Method]MTT assay and flow cytometry were used to detect the effects of different concentrations of diosgenindioscin on the proliferation and apoptosis of A549 cells.The enzyme activity test kit was used to detect cellular hexokinase(HK)and pyruvate kinase(PK)activities,and Western Blotting(WB)was applied to detect the expressions of proteins related to Hippo-YAP signaling pathway.[Result]MTT assay revealed that the IC50 of diosgenin on A549 cells at 24 h was 0.08μmol/L.With the increase of diosgenin concentration,the apoptosis rate of A549 cells increased in a dose-dependent manner.Compared with the control group,A549 cells in diosgenindioscinor Veteporfin-treated groups had reduced glucose uptake capacity and decreased lactate and ATP production(all P<0.05).Compared with the control group,A549 cells in diosgenindioscinor Veteporfin treatment groups showed reduced HK and PK activities(all P<0.05)and down-regulated protein expression levels of MOB1 and p-YAP(all P<0.05).[Conclusion]Diosgenin may interfere with the Hippo-YAP signaling pathway to inhibit glycolytic action and thereby promote apoptosis.
