摘 要:为了研究狐臭柴叶水提取液(Premna puberula Pamp.leaves water extract,PPW)对秀丽隐杆线虫的抗衰老作用,首先以秀丽隐杆线虫作为衰老模型,测定狐臭柴叶水提取液对秀丽隐杆线虫自然寿命、虫体长度、运动能力、紫外应激能力及丙二醛含量的影响,再以苯酚-硫酸法、酒石酸铁法、硝酸钠-硝酸铝法及考马斯亮蓝法分别测定PPW中多糖、总黄酮、总酚、蛋白质的含量。实验结果表明,与空白对照相比,在PPW含量为25%、50%、75%时,分别延长了秀丽隐杆线虫自然寿命的13.83%、16.42%、12.27%,在紫外应激条件下分别延长线虫寿命的21.12%、16.81%、32.96%。在PPW含量为50%和75%时,线虫的运动能力、虫体长度显著高于空白组(P<0.05),显著降低了线虫体内丙二醛的含量(P<0.05)。PPW中多糖、总黄酮、总酚、蛋白质含量分别为54.30±3.39、13.52±0.90、15.52±0.53、13.87±0.30 mg/g。结果说明,狐臭柴叶水提取液可以延缓秀丽隐杆线虫的衰老、降低体内丙二醛含量、促进线虫生长发育、提高运动能力和抗紫外应激能力。此结论可为狐臭柴叶进一步的研究与开发利用提供参考,尤其是为探究狐臭柴的食用价值、药用价值和医疗保健功能奠定理论基础。In order to study the anti-aging effect of Premna puberula Pamp.leaves water extract(PPW)on senescence of Caenorhabditis elegans.Using Caenorhabditis elegans as aging model,the effects of Premna puberula Pamp.leaves water extract on the natural life span,length of Caenorhabditis elegans,locomotor ability,UV stress ability and malondialdehyde content of Caenorhabditis elegans were studied.The contents of polysaccharide,total flavone,total phenol and protein in PPW were determined by phenol-sulfuric acid method,ferric tartrate method,sodium nitrate-aluminum nitrate method and Coomassie brilliant blue method.The experimental results showed that compared with the blank control,when the PPW content was 25%,50%,and 75%,the natural lifespan of Caenorhabditis elegans was extended by 13.83%,16.42%,and 12.27%,respectively.Under UV stress conditions,the lifespan of Caenorhabditis elegans was extended by 21.12%,16.81%,and 32.96%,respectively.When the PPW content was 50%and 75%,the locomotion ability and body length of Caenorhabditis elegans were significantly higher than those of blank group(P<0.05),and the malondialdehyde content in nematodes was significantly decreased(P<0.05).The contents of polysaccharide,total flavone,total phenol and protein in PPW were 54.30±3.39,13.52±0.90,15.52±0.53,13.87±0.30 mg/g,respectively.The results showed that the leaf water extract of Premna puberula Pamp.could delay the senescence of Caenorhabditis elegans,reduce the content of malondialdehyde,promote the growth and development of Caenorhabditis elegans and improve the ability of locomotion and anti-ultraviolet stress.The conclusion can be used as reference to the further research,development and utilization of the leaves of Premna puberula Pamp.,especially in exploring the edible value,medicinal value and medical care function of Premna puberula Pamp..
