摘 要:本研究旨在探讨过表达癌-睾丸抗原23(cancer-testis antigen 23,CT23)对肝细胞癌(hepatocellular carcinoma, HCC)细胞迁移、侵袭及上皮-间充质转化(epithelial-mesenchymal transition, EMT)的影响。基于在线数据库(TCGA、 GEO、 HPA)分析CT23在HCC组织和正常肝组织中的表达;建立稳定过表达CT23的人肝癌细胞Huh7,通过划痕实验和Transwell实验分别检测细胞迁移和侵袭能力,并观察细胞形态;采用基因集富集分析(gene set enrichment analysis, GSEA)探究CT23影响的通路及机制;利用免疫荧光检测纤维状肌动蛋白(filamentous actin, F-actin)分布;利用Western blot检测EMT标记蛋白表达。结果显示,CT23在HCC组织中高表达,并且CT23高表达在EMT和细胞骨架重塑过程显著富集。过表达CT23能促进细胞迁移和侵袭,呈现EMT形态学改变,EMT标志蛋白表达改变,如E-cadherin表达下调、 N-cadherin和Vimentin表达上调,以及F-actin组装加强。上述结果表明过表达CT23促进HCC细胞的迁移和侵袭,其机制可能与EMT和细胞骨架重塑相关。The purpose of this study wasto investigate the ffect of overexpression of cancer-testis antigen 23(CT23)on the migration,invasion and epithelial-mesenchymal transition(EMT)of hepatocellular carcinoma(HCC)cells.The expression of CT23 in HCC tissue and normal liver tissues was analyzed based on online database(TCGA,GEO,HPA).Stable overexpression of CT23 in hepatocellular carcinoma cell Huh7 was established.The migration and invasion ability of the cells were detected by wound healing assay and tran-swell assay.Gene set enrichment analysis(GSEA)was used to explore the pathways and mechanisms affected by CT23.Immunofluo-rescence was used to detect the filamentous actin(F-actin)distribution,and Western blot was used to detect the expression of EMT marker proteins.Bioinformatics analysis showed that CT23 was highly expressed in HCC tssues,and CT23 was significantly enriched during EMT and cytoskeleton remodeling.Overexpression of CT23 can promote cell migration and invasion,showing morphological chan ges of EMT,expression changes of EMT marker proteins,such as down-regulation of E-cadherin,and up-regulation of N-cadherin and Vimentin,and enhanced assembly changes of F-actin.These results suggest that overexpression of CT23 promotes the migration and inva-sion of HCC,and its mechanism may be related to EMT and cytoskeleton remodeling.
