摘  要:目的探讨肥胖易感性与肠道菌群及炎症反应的关系。方法32只雄性SD大鼠随机分为对照组(control,CON)8只,喂饲基础饲料;24只喂饲高脂饲料,8w后体重增量较高的前1/3为肥胖易感组(obesity-prone,OP),后1/3为肥胖抵抗组(obesity-resistant,OR)。麻醉处死后剥离大鼠内脏脂肪称重并计算脂体比,肝脏组织进行病理学检查;采集血清检测氧化应激、血生化和炎症细胞因子水平;利用16SrDNA测序技术分析肠道菌群的群落构成及多样性,并与炎症相关性分析;同时利用real-time PCR对常见菌株进行定量分析。结果OP组大鼠体重、内脏脂肪重、脂体比和血脂水平均高于CON组和OR组大鼠(P<0.05),OR组与CON组相比血脂水平未见明显改变;与CON组相比,OP和OR组大鼠均发生了不同程度的氧化损伤和抗氧化能力的减弱,并均发生了炎症反应;各组大鼠肠道菌群构成在门水平和属水平均表现出不同,肠道内优势菌属发生明显改变;炎症因子IL-4和IL-10水平与Bacteroidales_S24-7_group_n ank、Desulfovibrio含量呈负相关,与Lactobacillus含量呈正相关;促炎性细胞因子TNF-α水平与Lactobacillus含量呈负相关;且三组小鼠肠道中常见菌株Bifidobacteriumgenus在三组中CON组明显高于OP组和OR组(P<0.05);Bacteroidesgenus在OR组的含量是最高的,并明显高于OP组(P<0.05);Lactobacillusgenus在CON组的含量明显高于OP组、OR组(P<0.05)。结论肥胖易感性的不同与肠道菌群结构差异及炎症反应程度不同存在着相关性,并伴随氧化应激损伤、血脂水平的不同。Objective To explore the relationship between obesity susceptibility and intestinal flora and inflammatory response.Methods Thirty-two male SD rats were randomly divided into control group(CON,8 rats fed a basic diet).Twenty-four rats were fed a high-fat diet.After 8 weeks,the first third of the rats who gained more weight were obesity-prone(OP)and the last third were obesity-resistant(OR).After anesthesia,the visceral fat was stripped and weighed,and the ratio of fat to body was calculated.The liver tissue was examined pathologically.Serum was collected to detect oxidative stress,blood biochemistry and inflammatory cytokines.16S rDNA sequencing technology was used to analyze the community composition and diversity of intestinal flora,and the correlation with inflammation was analyzed.Meanwhile,real-time PCR was used for quantitative analysis of common strains.Results The body weight,visceral fat weight,fat-to-body ratio and blood lipid levels of the OP group were higher than those of the CON group and OR group(P<0.05),but there was no significant difference in blood lipid levels between the OR group and the CON group.Compared with the CON group,rats in the OP and OR groups had different degrees of oxidative damage,weakened antioxidant capacity and inflammatory reaction.The compositions of the intestinal flora in the three groups were different at phylum and genus leves and the dominant bacteria genera in the intestinal tract were also different.The levels of inflammatory factors IL-4 and IL-10 were negatively correlated with the contents of Bacteroidales_S24-7_group_n ank and Desulfovibrio,but positively correlated with the contents of Lactobacillus.The level of inflammatory cytokine TNF-ɑwas negatively correlated with the content of Lactobacillus.Bifidobacterium genus,a common strain in the intestinal tract of the three groups was significantly higher in the CON group than in the OP group and OR group(P<0.05).The content of Bacillus genus was the highest in the OR group(P<0.05).The content of Lactobacillus gen
