摘　　要：癌症作为全球最常见的死亡原因,其发病率和死亡率呈逐年上升趋势,严重危害人类健康。随着目前治疗方式的进步,部分患者的预后得到明显改善,但总体生存率仍然很低。本研究对胰岛素样生长因子结合蛋白2(IGFBP2)的结构、功能及其在肿瘤中的研究进展进行综述,以期为肿瘤的诊断和治疗提供新的方向和思路。以“IGFBP2”“肿瘤”“癌症”和“治疗”为关键词,检索2018-10-01-2022-09-30中国知网及PubMed数据库相关文献。纳入标准:IGFBP2的结构与功能;IGFBP2在癌症中的研究进展。排除标准:涉及IGFBP2的著作、汇编及二次文献;质量较差的文献。最终纳入分析文献45篇。结果表明,IGFBP2是相关性分泌蛋白家族中的一员。IGFBP2由N端结构域、连接子和C端结构域构成,其结构的特殊性使得其可以与整合素、肝素和高亲和力胰岛素样生长因子(IGF)结合,并在细胞增殖、分化以及人体生长发育中发挥重要的作用。IGFBP2在肺癌、胰腺癌以及结直肠癌等肿瘤的发生发展中发挥重要作用。IGFBP2可以通过整合多种信号通路、促进一些因子核积累、调控肿瘤微环境(TME)、保护肿瘤免受凋亡影响等方式来促进肿瘤细胞增殖、侵袭和迁移。IGFBP2还可以增加部分肿瘤的化疗耐药性,导致化疗失败。此外,IGFBP2高表达还与肿瘤的不良预后呈正相关。越来越多的学者将IGFBP2作为癌症治疗的新靶点进行研究并取得很大成就。但目前临床上尚无成熟的IGFBP2抑制剂投入应用。因此,探究IGFBP2在肿瘤中的发生机制及作用,寻找新的IGFBP2的相关靶点,对肿瘤的诊断与治疗具有深远意义。Cancer has become the most common cause of death in the world,and its incidence rate and mortality are increasing year by year,which has seriously endangered human health.With the progress of current treatment,the prognosis of some patients has improved significantly,but the overall survival rate is still very low.This article reviewed the structure,function and research progress of insulin-like growth factor binding protein 2(IGFBP2)in tumors,in order to provide new direction and ideas for tumor diagnosis and treatment.With"IGFBP2,tumor,cancer and treatment"as the keywords,we searched relevant literature in CNKI and Pubmed database from October 1,2018to September 30,2022.Inclusion criteria:structure and function of IGFBP2,research progress of IGFBP2in cancer.Exclusion criteria30.424.9:works,compilations and secondary literature involving IGFBP2,and low-quality literature.Totally 45articles were included in the analysis.The results showed that IGFBP2was a member of the family of related secretory proteins.IGFBP2consists of an N-terminal structural domain,a linker and a C-terminal structural domain,and its structural specificity allows it to bind to integrins,heparin and high-affinity insulin-like growth factor(IGF)and to play an important role in cell proliferation,differentiation,and human growth and development.IGFBP2plays an important role in the occurrence and development of lung cancer,pancreatic adenocarcinoma and colorectal cancer.IGFBP2can promote tumor cell proliferation,invasion,and migration by integrating multiple signaling pathways,promoting nuclear accumulation of some factors,regulating the tumor microenvironment(TME),and protecting tumors from apoptosis.IGFBP2can also increase chemotherapy resistance in some tumors,leading to chemotherapy failure.In addition,high IGFBP2expression is positively correlated with poor prognosis of tumors.More and more scholars are studying IGFBP2as a new target for cancer therapy and have achieved great success.However,there are no mature IGFBP2inhibitors in clinical
