摘  要:目的:研究新型肿瘤特异性外周血淋巴细胞(human periphral lymphosytals,HPL)对胃癌细胞的凋亡调控作用。方法:为制备一种新型肿瘤特异性HPL,我们首先分离出外周血中单个核细胞(peripheral blood mononuclear cells,PBMC),接着利用脂质体介导的细胞转染技术,将重组载体scFv-CD3ζ-pcDNA3.0导入PBMC,成功获得了目标细胞。通过对HGC27和SGC7901细胞使用新型肿瘤特异性HPL进行治疗,并采用Annexin V-FITC流式细胞术来检测细胞凋亡信号的表达程度。此外,通过免疫印迹法检测Bcl-2、Bcl-x和Bax凋亡蛋白在这两组胃癌细胞中的变化,以研究新型肿瘤特异性HPL对胃癌细胞凋亡的调控作用。结果:经使用新型肿瘤特异性HPL治疗胃癌HGC27和SGC7901细胞,并检测胃癌细胞凋亡信号Annexin V的表达情况,发现导致HGC27细胞早期和晚期凋亡率为(62.46±4.1)%,高于SGC7901细胞早期和晚期凋亡率(15.40±4.8)%(P<0.05)。通过免疫印迹法分析发现,HGC27细胞中的促凋亡蛋白Bax表达水平明显增加,而SGC7901细胞中Bax表达水平明显减少。此外,在HGC27细胞中,抗凋亡蛋白Bcl-2和Bcl-x的表达水平显著降低,而在SGC7901细胞中,它们的表达水平明显增加。结论:研究表明,新型肿瘤特异性HPL能够引发胃癌HGC27细胞的凋亡信号表达,增加凋亡蛋白Bax的表达,并抑制Bcl-2和Bcl-x的表达,从而诱发胃癌细胞凋亡。本研究结果有望实现对胃癌的有效治疗。Objective:To study the effect of new tumor-specific human peripheral lymphosytals(HPL)on apoptosis of gastric cancer cells.Methods:In order to prepare a novel tumor-specific HPL,we first isolated peripheral blood mononuclear cells(PBMC)from peripheral blood,and then introduced the recombinant vector scFv-CD3ζ-pcDNA3.0 into PBMC by liposome-mediated cell transfection technology,and successfully obtained the target cells.HGC27 and SGC7901 cells were treated with new tumor-specific HPL,and the expression of apoptosis signal was detected by Annexin V-FITC flow cytometry.In addition,the changes of Bcl-2,Bcl-x and Bax apoptosis proteins in these two groups of gastric cancer cells were detected by Western blot to study the regulatory effect of new tumor-specific HPL on gastric cancer cell apoptosis.Results:The new tumor-specific HPL was used to treat gastric cancer HGC27 and SGC7901 cells,and the expression of apoptosis signal Annexin V was detected.It was found that the early and late apoptosis rate of HGC27 cells was(62.46±4.1)%,which was higher than that of SGC7901 cells(15.40±4.8)%(P<0.05).By Western blot analysis,it was found that the expression level of apoptosis-promoting protein Bax in HGC27 cells increased significantly,while the expression level of Bax in SGC7901 cells decreased significantly.In addition,the expression levels of anti-apoptosis proteins Bcl-2 and Bcl-x decreased significantly in HGC27 cells,but increased significantly in SGC7901 cells.Conclusion:The novel tumor-specific HPL can induce apoptosis of gastric cancer HGC27 cells,increase the expression of apoptosis protein Bax,and inhibit the expression of Bcl-2 and Bcl-x,thus inducing apoptosis of gastric cancer cells.The results of this study are expected to achieve effective treatment of gastric cancer.
