摘 要:目的:通过研究甲状腺乳头状癌(papillary thyroid carcinoma,PTC)患者和健康人的血清外泌体miRNA在不同碘水平下的表达谱差异,探寻不受高碘影响可稳定诊断PTC的生物标志物。方法:收集PTC患者和健康人的血清样本,采用砷铈催化分光光度法测定血清碘,分别纳入高碘、正常碘的PTC患者各10例,高碘、正常碘的健康人各5例作为研究对象。然后从血清中抽提外泌体,从中提取miRNA并进行高通量测序,进一步做基因本体论(gene ontology,GO)富集分析和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路分析。结果:分别筛选高碘、正常碘组中PTC患者与健康人的差异表达miRNA,再取交集得到12个共同差异表达miRNA,显著富集到6个PTC相关通路。结论:与健康人对比,PTC患者中下调的miR-3158-3p、miR-186-5p,上调的miR-122-5p、miR-375-3p、miR-10a-5p、miR-10b-5p、miR-148a-3p、miR-223-5p、let-7d-3p、miR-651-5p,这10个miRNA可以作为在一般人群中区别PTC患者和健康人的稳定潜在生物标志物。Objective:To explore biomarkers that can stably diagnose papillary thyroid carcinoma(PTC)without the influence of high iodine,the expression profiles of serum exosomal miRNA in patients with PTC and healthy subjects at different iodine levels were studied.Methods:Serum samples from PTC patients and healthy people were collected,and serum iodine was determined by catalytic spectrophotometry.PTC patients with high iodine and normal iodine were 10 cases respectively,and healthy people with high iodine and normal iodine were 5 cases respectively.Then,exosome was extracted from serum.MiRNAs of exosome were extracted and performed by high-throughput sequencing.Bioinformatics analysis of target gene was done by method of gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG).Results:The differential-expressed miRNAs of PTC patients and healthy people in the high iodine and normal iodine groups were screened separately.They were intersected from these two groups to obtain 12 common differential-expressed miRNAs,which were significantly enriched to 6 pathways related to PTC.Conclusion:Compared with healthy people,miR-3158-3p and miR-186-5p are down-regulated in PTC patients,and miR-122-5p,miR-375-3p,miR-10a-5p,miR-10b-5p,miR-148a-3p,miR-223-5p,let-7d-3p,miR-651-5p are up-regulated.These 10 miRNAs can be used as stable potential biomarkers to distinguish PTC patients from healthy individuals in the general population.
