芸豆CBF基因家族鉴定及低温胁迫下的表达分析

摘　　要：CBF(C-repeat binding factor)是在植物响应低温胁迫中发挥重要作用的一种可被快速诱导的关键信号转录因子。本研究对芸豆(Phaseolus vulgaris)CBF基因家族进行了生物信息学分析并利用转录组数据(RNA-seq)和qRT-PCR对芸豆CBF基因在低温胁迫下的表达进行了检测。结果表明,在芸豆中共鉴定出7个PvCBFs,不均匀分布在5条染色体上;系统进化分析发现,芸豆CBF成员被分为4个亚组(Ⅰ~Ⅳ),与基因结构和保守基序分析结果相同;共线性分析发现,芸豆和拟南芥(Arabidopsis thaliana)CBF基因家族之间存在5对直系同源基因对,PvCBFs基因之间存在6对旁系同源基因对;启动子分析发现,PvCBFs含有多种参与植物生长发育、激素以及逆境响应的顺式元件;组织表达分析发现,PvCBFs基因具有明显的组织表达特异性;qRT-PCR结果显示,3个PvCBFs在低温胁迫下显著上调表达(P<0.05)。本研究可为后续PvCBFs耐低温功能分析提供参考依据。CBF(C-repeat binding factor)is a key transcription factor that can be rapidly induced and plays an important role in the response of plants to chilling stress.In this study,bioinformatics analysis of the common bean(Phaseolus vulgaris)CBF gene family and the expression of common bean CBF genes under chilling stress were examined using transcriptome data(RNA-seq)and qRT-PCR.The results showed that a total of 7 PvCBFs were identified in common bean,unevenly distributed on 5 chromosomes;phylogenetic analysis revealed that common bean CBF members were classified into 4 subgroups(Ⅰ~Ⅳ),which were identical to the results of gene structure and conserved motif analysis;covariance analysis revealed that there were 5 pairs of CBF gene families between common bean and Arabidopsis thaliana.The promoter analysis revealed that PvCBFs contained a variety of cis-elements involved in plant growth and development,hormone and stress response.The qRT-PCR results showed that 3 PvCBFs were significantly up-regulated under chilling stress(P<0.05).This study can provide a reference for subsequent study of the chilling tolerance function of PvCBF genes.
