摘 要:目的:分析ATP柠檬酸裂解酶(ACLY)和溶血磷脂酰胆碱酰基转移酶1(LPCAT1)在口腔鳞状细胞癌的表达与临床意义,进一步探讨其潜在功能。方法:运用TCGA数据库分析ACLY和LPCAT1在口腔鳞状细胞癌中的表达。采用免疫组织化学方法检测ACLY和LPCAT1在OSCC和癌旁正常组织的表达,并探讨其表达与患者临床病理特征和预后的关系。结果:TCGA数据库显示ACLY和LPCAT1在OSCC中高表达(P<0.05);免疫组化结果显示,ACLY和LPCAT1在OSCC中高表达(P<0.001),两者结果呈现一致表达。ACLY的表达量与OSCC的分化程度相关(P=0.009),LPCAT1的表达量与OSCC的分化程度相关(P<0.001),Spearman分析OSCC中ACLY和LPCAT1的表达呈正相关(r=0.449,P<0.001)。Kaplan-Meier生存分析表明ACLY和LPCAT1高表达患者生存时间短(P<0.05),Cox多因素分析显示LPCAT1的表达量是OSCC的独立预后因素(P<0.05)。结论:ACLY和LPCAT1在OSCC中高表达,且两者表达呈正相关,LPCAT1有望成为患者独立预后的代谢相关生物标志物。Objective:To analyze the expression and clinical significance of ATP-citrate lyase(ACLY)and Lysophosphatidylcholine Acyltransferase 1(LPCAT1)in Oral Squamous Cell Carcinoma(OSCC).Methods:TCGA database was used to analyze the expression of ACLY and LPCAT1 in oral squamous cell carcinoma.Immunohistochemistry was used to detect the expression of ACLY and LPCAT1 in OSCC and paracancerous normal tissues,explore the relationship between their expression and clinical pathological features and prognosis of patients.Results:TCGA database showed that high expression of ACLY and LPCAT1 in OSCC(<0.05);The results of immunohistochemistry showed that high expression of ACLY and LPCAT1 in OSCC(<0.001).The expression of ACLY was correlated with the differentiation degree of OSCC(=0.009).The expression of LPCAT1 was correlated with the differentiation degree of OSCC(<0.001).Spearman analysis showed that the expression of ACLY and LPCAT1 in OSCC was positively correlated(r=0.449,<0.001).Kaplan-Meier survival analysis showed that patients with high ACLY and LPCAT1 ze-xpression had shorter survival time(<0.05).Cox multivariate analysis showed that LPCAT1 expression level(<0.05)was an independent prognostic factor of OSCC.Conclusion:High expression of ACLY and LPCAT1 in OSCC and there is a positive correlation between them.LPCAT1 is expected to be a metabolic biomarker for independent prognosis of OSCC.
