摘 要:姜黄素是姜黄中重要的功能性物质,研究采用乙醇浸提法对泉州本土10个不同品种姜黄的总姜黄素含量进行测定。其中引种自广西壮族自治区防城港市的A2品种的总姜黄素含量最高,达8.134%。以A2姜黄品种为原料,选取乙醇体积分数、料液比、粉碎目数、超声时间、超声功率为影响因素进行单因素试验,以总姜黄素含量为评价指标,在单因素基础上,利用响应面法探究乙醇体积分数、粉碎目数及超声时间对总姜黄素提取率的影响并进行优化。结果表明,3个因素对总姜黄素提取率的影响依次为:粉碎目数>超声时间>乙醇体积分数;A2姜黄品种的最佳提取工艺是:乙醇体积分数68%、料液比1:40(g/mL)、粉碎目数30目、超声时间31.00 min、超声功率为250 W,此时总姜黄素的提取率最高,达到8.352%,与预测值8.386%相近,该工艺条件可靠有效。抗氧化试验表明,总姜黄素提取液对羟基自由基及DPPH自由基均有较强的清除能力和抗氧化能力。Curcumin is an important functional substance in Turmeric(Curcuma longa L).This study is to measure total curcumin of 10 different local varieties Turmeric in Quanzhou through ethanol extraction.Among them,A2 variety introduced from Fangchenggang City,Guangxi Zhuang Autonomous Region has the highest total curcumin content,up to 8.134%.With A2 Turmeric variety as raw material,ethanol concentration,solid-liquid ratio,particle size,ultrasonic time and ultrasonic power were selected as experimental factors to conduct a single factor experiment.Taking the total curcumin content as the evaluation index,on the basis of single factor,response surface methodology was used to explore the effects of ethanol concentration,particle size and ultrasonic time on the extraction rate of total curcumin and to optimize them.The results showed that the effects of the three factors on the extraction rate of total curcumin were as follows:particle size>ultrasonic time>ethanol concentration.The best extraction process of A2 variety is:When the ethanol concentration is 68%,the ratio of material to liquid is 1:40(g/mL),the particle size is 30 mesh,the ultrasonic time is 31.00 mins,and the ultrasonic power is 250 W,the extraction rate of total curcumin is the highest,reaching 8.352%,which is close to the predicted value of 8.386%.The process conditions are reliable and effective.The antioxidant test showed that total curcumin extract has strong scavenging activity on hydroxyl radical and DPPH radical and antioxidant activity.
