肌肉组织液对番鸭脂肪细胞增殖分化和脂质沉积的影响

摘　　要：为研究肌肉组织液对脂肪细胞增殖、分化和脂质沉积的影响,试验在体外培养的番鸭脂肪细胞中添加不同浓度的肌肉组织液,以模拟肌内脂肪的微环境。结果显示,添加10%肌肉组织液对体外培养的番鸭脂肪细胞的增殖、分化有显著抑制作用,导致成熟脂肪细胞中脂质沉积显著减少(P<0.05),同时显著降低脂肪合成相关基因,即脂肪酸合成酶基因(FASN)、过氧化物酶体增殖剂激活受体γ基因(PPARγ)、细胞核受体共激活因子1和3(SRC1、SRC3)基因表达水平(P<0.05),显著升高脂肪分解相关基因脂肪甘油三酯脂肪酶(ATGL)表达水平(P<0.05)。结果表明,在体外培养的番鸭脂肪细胞中添加10%肌肉组织液能够模拟肌内脂肪的微环境,显著抑制脂肪细胞的增殖、分化和脂质沉积。In order to study the effect of muscle tissue fluid on proliferation,differentiation and lipid deposition of adipo-cytes,Muscovy duck adipocytes were cultured in vitro with different concentrations of muscle tissue fluid to simulate the microen-vironment of intramuscular adipocytes.The results showed that the addition of 10%muscle tissue fluid significantly inhibited the proliferation and differentiation of Muscovy duck adipocytes cultured in vitro,and resulted in a significant reduction in lipid depo-sition in mature adipocytes(P<0.05).At the same time,the expression levels of lipid anabolism related genes,such as fatty acid synthase(FASN),peroxisome proliferators-activated receptors(PPARγ),nuclear receptor coactivator(SRC1)and(SRC3)were de-clined significantly(P<0.05),the expression level of adipolysis related gene adipose triglyceride lipase(ATGL)was increased sig-nificantly(P<0.05).This results indicated that the supplement of 10%muscle tissue fluid in Muscovy duck adipocytes culture could simulate the microenvironment of intramuscular fat and significantly inhibit the proliferation,differentiation and lipid depo-sition of mature adipocytes.
