摘 要:为探究陕西榆林地区鸡蛔虫单倍型多态性及遗传分化情况,并阐明其种系发育关系,试验以分离于榆林不同地区的30条鸡蛔虫为样品,采用PCR扩增其线粒体cox1与nad4基因,进行单倍型多态性、遗传分化与种系发育关系分析。结果显示:在所有的cox1基因(953 bp)序列中,共含30个变异位点(4个转换、26个颠换),17个单倍型(h=17,Hd=0.936),其基因分化系数(Gst)为-0.01285,遗传分化系数(Fst)为-0.02995,基因流(Nm)为11.53;在所有的nad4基因(876bp)序列中,共含29个变异位点(2个转换、27个颠换),16个单倍型(h=16,Hd=0.9356),其Gst为-0.01238,Fst为-0.0409,Nm为17.05;在所有的cox1+nad4基因(1829 bp)序列中,共含59个变异位点(6个转换、53个颠换),24个单倍型(h=24,Hd=0.977),其Gst为-0.0039,Fst为-0.0365,Nm为14.77;在所有cox1、nad4及cox1+nad4基因的Network图中,均未发现有单倍型聚集成簇的现象,构建的种系发育树中30条鸡蛔虫均聚集于一起,共同形成种系发育树的一个分支。研究表明,分离于陕西榆林地区的30条鸡蛔虫之间虽存在一定程度的基因变异,但其个体及单倍型之间具有较近的亲缘关系,尚未出现遗传分化。To study the haplotypes diversity and genetic differentiation of Ascaridia galli(A.galli)collected from Yulin ar-ea,Shaanxi province,and to define its phylogenetic relationships,a total of 30 strains A.galli collected from Yulin area were used as the experiment samples.The mitochondrial genes cox1 and nad4 of A.galli were amplified by RCR,then sequenced and ana-lyzed.The results showed that there were 30 mutation sites that includes 4 bases transition sites and 26 bases transversion sites in all cox1 gene sequences;A total of 17 haplotypes(h=17,Hd=0.936)were found in cox1 gene sequences,and their coefficient of gene differentiation(Gst),coefficient of genetic differentiation(Fst)and gene flow(Nm)were-0.01285,-0.02995 and 11.53,re-spectively;Within all nad4 gene sequences,there were 29 mutation sites including 2 bases transition sites and 27 bases transver-sion sites,and total of 16 haplotypes(h=16,Hd=0.9356)were found and the values of Gst,Fst and Nm were-0.01238,-0.0409 and 17.05,respectively;There were 59 mutation sites which includes 6 bases transition sites and 53 bases transversion sites in all cox1+nad4 gene sequences;A total of 24 haplotypes(h=24,Hd=0.977)were found in cox1+nad4 gene sequences,and their Gst,Fst and Nm were-0.0039,-0.0365 and 14.77,respectively.Moreover,the phylogenetic analysis results showed that 30 strains A.galli collected from Yulin area together formed one of clades in each tree.The results indicated that 30 strains of A.galli isolated from Yulin area,Shaanxi province had some genetic variation,but their individuals and haplotypes had closer genet-ic relationships,and there was no genetic differentiation among individuals.
