摘  要:目的:研究迷走神经刺激(VNS)对慢性不可预知性温和应激(CUMS)模型大鼠抑郁样行为的影响。方法:利用CUMS方法制备大鼠抑郁模型,用VNS对大鼠进行治疗,糖水偏好实验和强迫游泳实验检测大鼠的抑郁样行为,高效液相色谱法(HPLC)检测大鼠脑脊液中5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)及去甲肾上腺素(NE)的含量。结果:与正常大鼠相比,CUMS组大鼠体重增加缓慢(P<0.05),糖水偏好率降低(P<0.05)且游泳不动时间增加(P<0.05),表现出明显的抑郁样行为,同时其脑脊液中5-HT、5-HIAA和NE含量显著降低(P<0.05)。经过VNS治疗,大鼠的体重增加明显(P<0.05),糖水偏好率增加(P<0.05)且游泳不动时间减少(P<0.05),与之相适应的是脑脊液中5-HT、5-HIAA和NE水平显著升高(P<0.05)。结论:VNS能够改善CUMS大鼠的抑郁样行为并提高脑脊液中5-HT和NE的水平。Objective:To study the effects of vagus nerve stimulation(VNS)on depression-like behavior in chronic unpredictable mild stress(CUMS)model rats.Methods:CUMS method was used to prepare the rat model of depression,and the rats were treated with VNS.The depression-like behavior of the rats was detected by sucrose preference test(SPT)and forced swimming test(FST),and the contents of 5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA),and norepinephrine(NE)in the cerebrospinal fluid of the rats were detected by high performance liquid chromatography(HPLC).Results:Compared with the control rats,the CUMS group showed a slow weight gain(P<0.05),a lower percentage of sucrose preference(P<0.05),an increased swimming immobility time(P<0.05),and a significant depression-like behavior.At the same time,5-HT,5-HIAA,and NE in the cerebrospinal fluid were significantly decreased(P<0.05).After VNS treatment,the body weight of rats increased significantly(P<0.05).The percentage of sucrose preference of the rats was increased(P<0.05)and the swimming immobility time was decreased(P<0.05).The 5-HT,5-HIAA,and NE levels in the cerebrospinal fluid were significantly increased(P<0.05).Conclusion:VNS can improve depression-like behavior and increase the levels of 5-HT and NE in cerebrospinal fluid of CUMS rats.
