摘 要:[目的]研究猫豆提取物对α-葡萄糖苷酶活性的抑制作用,并筛选出相关化合物。[方法]用系统溶剂提取法制备猫豆根、茎、叶、花、豆和豆荚的提取物,分别进行α-葡萄糖苷酶抑制实验,确定活性部位后,用液相-高分辨质谱分析各提取物中的化学成分,筛选出在活性部位高表达的化合物。[结果]猫豆根的正丁醇提取部位,叶的水提物Ⅰ和甲醇、正丁醇、氯仿提取部位,豆荚的甲醇、正丁醇和氯仿提取部位,对α-葡萄糖苷酶的半数抑制浓度(IC50)均小于10μg/ml,对α-葡萄糖苷酶具有较强的抑制作用。经质谱分析筛选出13个高表达的化合物:山柰酚-3-O-β-D-槐糖苷、山柰酚-3-O-鼠李半乳糖苷、7-羟基-6,4’-二甲氧基异黄酮、大豆异黄酮、花旗松素、柚皮苷查尔酮、高丽槐素、mucunone A、3-hydroxy-2-methyl-2,3-dihydrobenzofuran-4-carboxylic acid、新喋呤、3-甲基吲哚、2’-hydroxy-biochanin A、丁二酸,这些化合物多分布在正丁醇、甲醇及氯仿提取部位。[结论]从猫豆提取物中共筛选出13个对α-葡萄糖苷酶活性具有明显抑制作用的化合物,可作为进一步研究的物质基础。[Objective]To study the inhibitory effect of feline bean extract onα-glucosidase activity and screen out related compounds.[Methods]The extracts of roots,stems,leaves,flowers,beans and pods of Mucuna pruriens.were prepared by systematic solvent extraction method,and theα-glucosidase inhibition experiments were carried out respectively.After the active sites were determined,the chemical components of each extract were analyzed by liquidhigh resolution mass spectrometry,and the compounds with high expression in the active sites were screened.[Results]The n-butanol part of the root,the water fractionⅠof the leaves,methanol,n-butanol,trichloromethane part,and the methanol,n-butanol,trichloromethane part of the pod had the IC50 all less than 10μg/ml,which had a strong inhibitory effect onα-glucosidase.Thirteen highly expressed compounds were screened by mass spectrometry analysis,and the results were identified by comparing with the standard compound library as kaempferol-3-O-β-D-sophoroside,kaempferol-3-O-rhamnoside,7-hydroxy-6,4'-dimethoxyisoflavone,soybean isoflavone,taxifolin,naringin chalcone,maackiain,mucunone A,3-hydroxy-2-methyl-2.3-dihydrobenzofuran-4-carboxylic acid,neopterin,3-methylindole,2'-hydroxy-biochanin A,succinic acid.These compounds are mostly distributed in the n-butanol,methanol and chloroform extracts.[Conclusion]There were 13 compounds were screened out from Mucuna pruriens showed inhibility toα-glucosidase as the potential compounds for further studies.
