摘  要:目的:基于Kisspeptin/GPR54(吻素/吻素受体)通路研究二陈汤对高脂饮食小鼠生殖激素分泌的影响。方法:将30只C57BL/6J雌性小鼠随机分为正常组、模型组、二陈汤组,每组10只。模型组、二陈汤组小鼠给予高脂饲料喂养12周,第9周开始二陈汤组给予二陈汤灌胃,正常组和模型组小鼠予生理盐水灌胃,干预4周。记录小鼠基础数据,行葡萄糖耐量实验,检测血脂四项;实施HE染色观察卵巢形态,应用ELISA检测小鼠促卵泡激素(FSH)、促黄体生成素(LH)、睾酮(T)和雌二醇(E2)的水平;应用q-PCR与Western Blot检测小鼠下丘脑瘦素(Leptin)、Kisspeptin、GPR54、促性腺激素释放激素(GnRH)的表达水平。结果:与正常组比较,模型组小鼠体质量和腹围有较大幅度增长(P<0.05,P<0.01),血糖血脂水平升高(P<0.05,P<0.01),卵巢颗粒细胞排列松散,脂滴空泡明显增多,血清T、FSH和LH水平升高(P<0.05,P<0.01),下丘脑组织Leptin、Kisspeptin、GnRH的mRNA和蛋白的表达水平显著升高(P<0.01);与模型组比较,二陈汤组小鼠的体质量、腹围下降(P<0.05,P<0.01),血糖血脂水平下降(P<0.05,P<0.01),卵巢形态有所恢复,血清T、LH水平下降(P<0.05,P<0.01),下丘脑组织Leptin、Kisspeptin的mRNA表达水平显著降低(P<0.01),Kisspeptin的蛋白表达水平显著降低(P<0.05)。结论:二陈汤可能通过Kisspeptin/GPR54通路改善高脂饮食小鼠的血糖血脂水平,调节生殖激素分泌水平,改善小鼠卵巢组织解剖学与形态学特征。Objective:To study the effects of Erchen Decoction on glucose and lipid metabolism and reproductive hormone secretion in mice fed with high-fat diet based on Kisspeptin/GPR54 pathway.Methods:Thirty C57BL/6J female mice were randomLy divided into normal group,model group and Erchentang group,with 10 mice in each group.The mice in the model group and Erchen decoction group were fed with high-fat feed for 12 weeks,the mice in the Erchen decoction group were given Erchen decoction by gavage from the 9th week,and the mice in the normal group and model group were given normal saline by gavage for 4 weeks.Record the basic data of mice;Glucose tolerance test was performed to detect four items of blood lipids;Ovarian morphology was observed by HE staining,and FSH,LH,T and E_2 were detected by ELISA;The expression levels of leptin,Kisspeptin,GPR54 and GnRH in mouse hypothalamus were detected by q-PCR and Western blot.Results:Compared with the normal group,the body weight and abdominal circumference of mice in the model group increased(P<0.05,P<0.01);Ovarian granulosa cells were loosely arranged,lipid droplets and vacuoles were significantly increased,and serum levels of T,FSH and LH were increased(P<0.05,P<0.01),The mRNA and protein expression levels of leptin,Kisspeptin and GnRH in hypothalamus tissue were significantly increased(P<0.01);Compared with the model group,the body weight and abdominal circumference of the mice in the Erchentang group decreased(P<0.05,P<0.01),The level of blood glucose and blood lipid decreased(P<0.05,P<0.01),Ovarian morphology recovered,and serum T and LH levels decreased(P<0.05,P<0.01),The mRNA expression level of leptin and Kisspeptin in hypothalamus tissue was significantly decreased(P<0.01),and the protein expression level of Kisspeptin was significantly decreased(P<0.05).Conclusion:Erchen Decoction may regulate blood glucose,blood lipid level and reproductive hormone secretion level of mice fed with high-fat diet through Kisspeptin/GPR54 pathway,and improve the anatomical and morphologica
