摘 要:目的:优化川芎油Ligusticum chuanxiong Hort.oil(CXO)的提取工艺并研究其抗氧化作用。方法:在筛选CXO提取方法基础上,采用Box-Behnken响应面法优化其提取工艺,得到最佳提取条件。通过研究CXO对1,1-二苯基-2-苦酰肼(DPPH)、O^(2-)·的清除能力,探讨其抗氧化作用。结果:乙醚-超声萃取法作为最佳提取工艺,当川芎的粒度为中等粉末,烘烤温度为40℃,料液比为1∶10时,提取率最高。此时CXO提取率的预测值可达7.13%,实际值可达7.07%。此外,发现CXO以剂量依赖的方式清除DPPH和O^(2-)·,显示出强大的抗氧化能力。结论:通过乙醚-超声萃取法能够大幅提高CXO的提取率,所提取的CXO具有较强的抗氧化能力,解决了提取效率低和资源浪费严重的问题,为CXO在食品和医药方面的进一步开发利用提供科学依据。Objective:To optimize the extraction process of Ligusticum chuanxiong Hort.oil(CXO)and to study its antioxidant effect.Methods:Based on the screening of the CXO extraction method,the Box-Behnken response surface method was used to optimize its extraction process and obtain the optimal extraction conditions.The antioxidant effect of CXO was investigated by studying the scavenging ability of CXO on 1,1-diphenyl-2-picrylhydrazide(DPPH)and O^(2-)·.Results:Ether-ultrasonic extraction was used as the optimal extraction process,and the highest extraction rate was obtained when the particle size of CX was medium powder,the roasting temperature was 40℃,and the material-liquid ratio was 1∶10.At this time,the predicted value of CXO extraction rate could reach 7.13%and the actual value could reach 7.07%.In addition,CXO was found to scavenge DPPH and O^(2-)·in a dose-dependent manner,showing a strong antioxidant capacity.Conclusion:The extraction rate of CXO can be greatly improved by EUE,and the extracted CXO has strong antioxidant capacity.It solves the problems of low extraction efficiency and serious resource waste,and provides the scientific basis for the further development and utilization of CXO in food and medicine.
