摘 要:目的探讨脑胶质瘤患者血清长链非编码核糖核酸(long noncoding RNA,lncRNA)核富集转录体(nuclear enriched abundant transcript 1,NEAT1)表达与放疗敏感性的关系。方法选取96例脑胶质瘤患者作为研究对象,患者均进行标准根治术放疗治疗。放疗完成后,参照世界贸易组织(WTO)的实体瘤疗效评定标准(RECIST)将其分为放疗敏感组和放疗耐受组。收集患者年龄、性别、病理分级、肿瘤直径、肿瘤位置、肿瘤数目等临床资料;放疗前采用RT-PCR检测患者血清lnc RNA NEAT1表达水平;采用Logistic回归分析脑胶质瘤患者发生放疗耐受的影响因素。结果96例患者中放疗敏感78例。放疗敏感组病理分级Ⅰ~Ⅱ级患者(42.31%)多于放疗耐受组(16.67%)(χ^(2)=4.103,P=0.043),2组年龄、性别、体重指数、肿瘤直径、肿瘤位置、肿瘤数目比较,差异无统计学意义(P>0.05);放疗敏感组血清lnc RNA NEAT1为(2.04±0.69),低于放疗耐受组的(3.21±1.05),差异有统计学意义(t=5.828,P<0.05)。血清lnc RNA NEAT1(OR=1.086,95%CI:1.015~1.162,P<0.05)是脑胶质瘤患者放疗耐受的影响因素。结论脑胶质瘤患者血清lnc RNA NEAT1表达与放疗敏感性密切相关,血清lnc RNA NEAT1高表达是脑胶质瘤患者发生放疗耐受的危险因素。Objective To investigate the relationship between serum long noncoding RNA(lncRNA)nuclear enriched transcript 1(NEAT1)expression and radiosensitivity in patients with glioma.Methods 96 patients with glioma were treated with standard radical radiotherapy.After completion of radiotherapy,they were divided into radiosensitive and radioresistant groups according to the World Trade Organization(WTO)Response Evaluation Criteria in Solid Tumors(RECIST).Clinical data such as age,gender,pathological grade,tumor diameter,tumor location,and tumor number were collected;serum lnc RNA NEAT1 expression levels were measured by RT-PCR before radiotherapy;and logistic regression was used to analyze the influencing factors of radiotherapy tolerance in glioma patients.Results Of the 96 patients,78 were radiosensitive.The patients with pathological gradeⅠ~Ⅱin the radiosensitive group(42.31%)was more than that in the radiosensitive group(16.67%)(χ^(2)=4.103,P=0.043),and there was no significant difference in age,gender,body mass index,tumor diameter,tumor location,and tumor number between the 2 groups(P>0.05);serum lnc RNA NEAT1 was(2.04±0.69)in the radiosensitive group,which was lower than(3.21±1.05)in the radiosensitive group,(t=5.828,P<0.05).Serum lnc RNA NEAT1(OR=1.086,95%CI:1.015~1.162,P<0.05)was a factor influencing radiotherapy tolerance in glioma patients.Conclusion The expression of serum lnc RNA NEAT1 is closely related to radiosensitivity in patients with glioma,and the high expression of serum lnc RNA NEAT1 is a risk factor for radio resistance in patients with glioma.
